摘要
【目的】研究马铃薯提取物糖苷生物碱对化疗性静脉炎兔模型白介素-1(IL-1)、白介素-6(IL-6)、肿瘤坏死因子(TNF-α)表达的影响,探讨其减轻化疗性静脉炎的作用机理,为临床应用提供依据.【方法】将60只普通级‘日本大耳白兔’随机分为6组:空白对照组,模型组,阳性对照组,糖苷生物碱组,马铃薯汁组,马铃薯泥组.通过耳缘静脉推注长春瑞滨建立静脉炎模型,各试验组在推注长春瑞滨后在穿刺部位涂抹相应药物,免疫组化法检测IL-1、IL-6的含量,ELISA法检测各组兔血清中TNF-α的含量.【结果】IL-1、IL-6的表达:模型组含量与空白对照组比较显著增加,差异有统计学意义,与模型组比较,阳性对照组及各药物组IL-1、IL-6表达均有所减弱(P<0.05,P<0.01),以阳性对照组,糖苷生物碱组减弱最为明显;TNF-α含量表达:与空白对照组比较,模型组血清中TNF-α含量表达异常增高,与模型组比较,除马铃薯汁组外,阳性对照组、马铃薯泥组、糖苷生物碱组TNF-α表达均有所减弱(P<0.05,P<0.01).【结论】马铃薯糖苷生物碱对化疗性静脉炎家兔促炎细胞因子IL-1、IL-6、TNF-α的异常升高有显著的回调作用.
【Objective】To study the effect of glycoside alkaloid of potato extract on the expression of interleukin-1(IL-1),interleukin-6(IL-6)and tumor necrosis factor(TNF-α)in the rabbit model of chemotherapeutic phlebitis,and to explore the mechanism of alleviating chemotherapeutic phlebitis and provide a basis for clinical application.【Method】60 ordinary Japanese white rabbit(half male and half female)were randomly divided into 6 groups:control group,model group,positive control group,glucoside alkaloid group,potato juice group and potato slime group.The phlebitis model of Changchun Rurbin was injected into the ear vein.The experimental groups were applied to the puncture site after the injection of Changchun ribin(vinorelbine).The contents of IL-1 and IL-6 were detected by immunohistochemistry.The content of TNF-αin the serum of each group was detected by ELISA.【Result】Expression of IL-1 and IL-6:The content of the model group was significantly increased compared with the control group,and the difference was statistically significant.Compared with the model group,the expression of IL-1 and IL-6 in the positive control group and the drug applied groups were decreased(P<0.05,P<0.01),and the most obvious decrease in the glycoside alkaloid group was found in the positive control group.The expression of TNF-αcontent:Compared with the blank control group,the expression of TNF-αin the serum of model group was abnormally increased.Compared with the model group,the expression of TNF-αin positive control group,potato slime group and glycoside alkaloid group decreased(P<0.05,P<0.01).【Conclusion】Potato glycoalkaloids showed significant callback effects on the abnormal elevation of proinflammatory cytokines IL-1,IL-6 and TNF-αin rabbits with chemotherapy-induced phlebitis.
