摘要
目的研究不同类型环氧化酶抑制剂对急性芬太尼诱导的大鼠痛觉敏化的抑制作用。方法将40只SD雌性大鼠随机分成5组(n=8),即生理盐水组(NS组)、芬太尼组(F组)、帕瑞昔布钠组(P组)、氯诺昔康组(L组)及酮咯酸氨丁三醇组(T组)。除NS组外,其余各组皮下注射芬太尼50μg/kg×4次,间隔时间15 min。P组、L组、T组分别于注射芬太尼前15 min给予腹腔注射帕瑞昔布钠10 mg/kg、氯诺昔康0.5 mg/kg、酮咯酸氨丁三醇5 mg/kg。NS组全程注射生理盐水。在给药前1 d(基础)、注射芬太尼后1 h(1 h)和第1~4天(1~4 d)分别进行智能热板仪测试测量热痛阈值。结果皮下注射芬太尼可引起大鼠的热痛觉敏化;1、2 d,P、L及T组的热痛阈值均高于F组(P<0.05);3 d,P及T组的热痛阈值均高于F组(P<0.05);4 d,T组热痛阈值高于F组(P<0.05)。结论不同类型的环氧化酶抑制剂均对芬太尼诱导痛觉敏化大鼠的热痛阈值的降低有抑制作用,且酮咯酸的作用时间更长。
Objective To investigate the inhibited effects of different kinds of cyclooxygenase inhibitors in acute fentanyl induced hyperalgesia in rats. Methods Forty female rats were divided into 5 groups (with 8 cases in each group), including normal saline group (NS group), fentanyl group (F group), parecoxib sodium group (P group), lornoxicam group (L group) and ketorolac tromethamine group (T group). Except NS group, the other groups were subcutaneously injected with fentanyl 50 μg/kg×4 times, with an interval of 15 minutes. The groups P, L and T were given parecoxib sodium(10 mg/kg), lornoxicam(0.5 mg/kg) and ketorolac tromethamine(5 mg/kg) respectively at 15 minutes before fentanyl injection. The NS group injected saline throughout the course. The threshold of heat pain was measured by intelligent hot plate instrument at 1 day before administration(baseline), 1 hour after fentanyl injection (1 h) and 1-4 days(1-4 d). Results Subcutaneous injection of fentanyl can induce hyperalgesia in rats. On 1 and 2 days, the threshold of heat pain in the group P, L and T was higher than that in the group F(P<0.05). On day 3, the threshold of heat pain in the group P and T was higher than that in the group F (P<0.05). On day 4, the threshold of heat pain in the group T was higher than that in the group F(P<0.05). Conclusion Different kinds of cyclooxygenase inhibitors can get inhibited effects on the decrease of heat pain threshold of rats with acute fentanyl induced thermal hyperalgesia, and ketorolac had a longer effect.
