小檗碱/IGF-I对猪体外受精胚胎发育作用初探 被引量:2

摘要 【目的】初步探讨小檗碱(Berberine,Ber)和胰岛素样生长因子I (Insulin like growth factorI,IGF-I)对猪体外受精胚胎发育效果的作用。【方法】通过在NCSU-23基础培养液中添加IGF-I (50、100、150ng/mL),筛选出最佳质量浓度;以添加Ber (0.1μg/mL,Ber组)、最佳质量浓度的IGF-I (IGF-I组,100ng/mL)和同时添加二者(Ber+IGF-I组)为三个试验组,观察猪体外受精胚胎的发育效果,并检测各组别IGF-I、IGF-IR(Insulin-like growth factor I recepter,胰岛素样生长因子I受体)转录水平。【结果】IGF-I 100、150ng/mL组胚胎各时期发育率均极显著高于对照组和50ng/mL组(P<0.01),100ng/mL与150ng/mL组之间无显著差异(P>0.05),最佳质量浓度为100ng/mL;Ber组、IGF-I组、Ber+IGF-I组胚胎各时期发育率、IGF-I、IGF-IR转录水平均极显著高于对照组(P<0.01),但三组之间均无显著差异(P>0.05)。【结论】Ber和IGF-I可显著提高猪体外受精胚胎发育率,提高其内源性IGF-I、IGF-IR基因转录水平,但同时添加二者与单独添加相比并无显著变化,推测二者可能参与介导同一个信号通路,有待进一步研究。 【Objective】To investigate the effects of Berberine(Ber)and Insulin-like Growth Factor I(IGF-I)on the development of porcine embryos after in vitro fertilization.【Methods】The optimum concentration of IGF-I was screened by adding(50,100,150ng/mL)to the basic culture medium of NCSU-23.Using Ber(0.1μg/mL,Ber group),the optimum concentration of IGF-I(100μg/mL,IGF-I group)and both of them(Ber+IGF-I group)as experimental groups,the development effect of porcine IVF embryos was observed.The transcription level of IGF-I,Insulin-like Growth Factor I recepter(IGF-IR)was detected in all groups.【Results】The rates of embryonic development in IGF-I 100and 150ng/mL groups were significantly higher than that in control group and 50ng/mL group(P<0.01)and there was no significant difference between 150ng/mL group and IGF-I 100ng/mL(P>0.05).The optimum concentration was 100ng/mL.The rates of embryonic development and transcription levels of IGF-I and IGF-IR of embryo in Ber、IGF-I and Ber+IGF-I groups were significantly higher than that in control group(P<0.01),but there was no significant difference among the three groups(P>0.05).【Conclusion】Ber and IGF-I can significantly increase the rate of in vitro fertilization embryo development of pig and the transcription levels of IGF-I,IGFIR gene were increased,but the addition of both had no significant difference compared with the addition alone.It is inferred that they may be involved in mediating the same signal pathway,which should be further studied.
