摘要
目的:探究注射外源性胰岛素对限饲小鼠血糖、肝糖原、肌糖原含量的影响。方法:40只昆明种小鼠,随机均分4组(Ⅰ~Ⅳ组),分别为对照组、不限饲组、低限饲组和中限饲组,其中Ⅰ组皮下注射等量无菌水,自由采食;Ⅱ组皮下注射胰岛素1U,自由采食;Ⅲ组皮下注射胰岛素1U,供给正常采食量的80%日粮;Ⅳ组皮下注射胰岛素1U,供给正常采食量的60%日粮。各组均每天给药1次,试验期14d,试验期结束时禁食12h,不禁水,摘眼球采血,制备血清,处死小鼠,肝左叶腹面中部、胸部前锯肌、左腿部股二头肌采样,分别用于血糖,肝、肌糖原含量测定。结果:血糖水平:与Ⅱ组比较,Ⅲ组血糖水平降低(P<0.05),Ⅳ组血糖水平显著降低(P<0.01)。肝糖原含量:与Ⅰ组比较,Ⅱ组升高(P<0.05);与Ⅱ组比较,Ⅲ、Ⅳ组降低(P<0.01)。前锯肌糖原含量:与Ⅰ组比较,Ⅲ、Ⅳ组升高(P<0.01);与Ⅱ组比较,Ⅲ、Ⅳ组升高(P<0.01),且Ⅳ组高于Ⅲ组(P<0.05)。股二头肌糖原含量:与Ⅰ组相比,Ⅱ组升高(P<0.05),Ⅲ、Ⅳ组升高(P<0.01);与Ⅱ组比较,Ⅳ组升高(P<0.05)。结论:胰岛素可以降低小鼠的血糖,提高其肝糖原含量;胰岛素联合饲料控制可降低血糖和肝糖原含量的同时提升肌糖原含量。
Objective:To investigate the effect of injection of exogenous insulin on blood glucose,liver glycogen and muscle glycogen concentration of limited feeding mice.Methods:Forty Kunming mice were randomly divided into 4 groups.Groups I to IV were control group(subcutaneous injection of equal volume of sterile water,adlibitum feeding),no feeding group(subcutaneous injection of insulin 1 U,free mining food),low-limited diet group(subcutaneous injection of insulin 1 U,80%of the normal feed intake)and medium-limited group(subcutaneous injection of insulin 1 U,60%of the normal feed intake),daily once administered.The test period was 14 days.At the end of the test,the mice were fasted for 12 hours.The water was removed,the blood was collected from the eyeball,the serum was prepared,the mice were sacrificed,the ventral surface of the left lobe of the liver,the anterior serratus of the chest,and the biceps femoris of the left leg were sampled.for the determination of blood sugar,liver and muscle glycogen concentration.Results:Compared with groupⅡ,the blood glucose level in groupⅢwas decreased(P<0.05),and the blood glucose level in groupⅣwas significantly lower(P<0.01).Compared with the groupⅠ,the hepatic glycogen content in groupⅡincreased(P<0.05);compared with the groupⅡ,the groupsⅢ,Ⅳdecreased(P<0.01).The content of glycogen in anterior serratus was higher than that in the groupⅠ(P<0.01).Compared with the no-feed groupⅡ,the groupsⅢ,Ⅳwere higher(P<0.01).The groupⅣwas higher than groupⅢ(P<0.05).The concentration of glycogen in the biceps femoral muscle was higher than that in groupⅠ(P<0.05),and increased in groupsⅢⅣ(P<0.01).Compared with groupⅡ,the groupⅣwas elevated(P<0.05).Conclusion:Insulin can reduce blood sugar and increase liver glycogen content in mice.Insulin combined feed control can reduce blood sugar and liver glycogen concentration while increasing muscle glycogen concentration.
