香附四物汤中阿魏酸和芍药苷的大鼠肠吸收动力学研究 被引量:3

摘要 目的:研究香附四物汤中阿魏酸和芍药苷的大鼠肠吸收动力学特征。方法:通过建立大鼠在体单向肠灌流模型,以酚红为标示物,采用高效液相色谱法测定大鼠肠灌流液中阿魏酸和芍药苷的质量浓度,紫外分光光度计法测定肠灌流液中酚红即时浓度,考察不同肠段、不同药物浓度对香附四物汤中阿魏酸及芍药苷的吸收规律影响。结果:阿魏酸、芍药苷在pH 5.51~8.98的Krebs-Ringer(K-R)液中稳定性良好。不同浓度下的芍药苷或阿魏酸在大鼠各个不同肠段中的Peff值均无显著性差异,表明了芍药苷、阿魏酸在大鼠小肠内的吸收无自身浓度抑制作用,且主要以被动转运方式吸收。阿魏酸、芍药苷在大鼠四个肠段(十二指肠、空肠、回肠、结肠)的吸收速率参数值(Ka)、有效渗透系数值(Peff),都表明了阿魏酸、芍药苷在大鼠整个肠道内均有一定的吸收,且无特定的吸收部位。结论:香附四物汤中芍药苷、阿魏酸在大鼠小肠中较易吸收,吸收方式以被动转运为主,无特定吸收部位。 Objective:To investigate the intestinal absorption characteristic of ferulic acid and paeoniflorin in Xiangfu Siwu Tang.Methods:A one-way intestinal perfusion model in situ was established,with phenol red as a marker.The phenol red intestinal perfusate was characterized by UV,and ferulic acid and paeoniflorin were detected by HPLC.The in situ of ferulic acid and paeoniflorin were investigated at different concentrations and in different intestinal,respectively.Results:The stability of ferulic acid and paeoniflorin was excellent in Krebs-Ringer(K-R)solution when pH value was 5.51-8.98.No significant difference of P eff value was found in different concentrations of ferulic acid and paeoniflorinin different intestine segments,which showed that the absorption of ferulic acid and paeoniflorin was mainly complied with the passive diffusion.The P eff and K a values of intestinal could be absorbed in perfusion in duodenum,jejunum,ileum and colon which showed that ferulic acid and paeoniflorin have absorption in the whole intestine and there is no special absorption site.Conclusion:Ferulic acid and paeoniflorin were absorbed in the whole intestine where there is no special absorption site.Additionally,the intestine absorption mechanism of ferulic acid and paeoniflorin in Xiangfu Siwu Tang was passive diffusion.
