温阳消癥方对单侧输尿管梗阻小鼠肾间质纤维化的影响

Effect of Wenyangxiaozheng Decoction on Renal Interstitial Fibrosis in Mice with Unilater Ureteral Obstruction

目的:研究温阳消癥方对单侧输尿管结扎小鼠肾功能及肾组织病理的影响。方法:32只C57小鼠随机抽取8只作为假手术组,其余24只采用单侧输尿管结扎法制作肾间质纤维化模型。造模成功后随机分为模型组、缬沙坦组、温阳消癥方组,每组8只,分组给药,治疗2周后眼眶采血并测定血清肌酐、尿素氮,同时取肾组织进行HE、Masson染色,观察肾脏病理改变。结果:与假手术组相比,模型组的血清肌酐、尿素氮水平明显升高(P <0. 001),肾间质纤维化改变明显,胶原染色阳性率百分比升高(P <0. 001);与模型组比较,缬沙坦组和温阳消癥方组的血清肌酐、尿素氮水平均有明显下降(P <0. 001),肾组织纤维化程度较轻,胶原染色阳性率百分比下降(P <0. 001);温阳消癥方组疗效优于缬沙坦组。结论:温阳消癥方能改善UUO小鼠的肾功能,减轻肾间质纤维化,其效果好于缬沙坦胶囊。

Objective: To investigate the effect of Wenyangxiaozheng decoction on renal interstitial fibrosis in mice with unilater ureteral obstruction( UUO). Methods: 8 out of 32 C57 mice were randomly chosen to treated as sham operation group. The other24 mice established UUO mouse modle with renal interstitial fibrosis,then they were randomly divided into three groups: model group,valsartan group and WYXZ group,n = 8 in each group. Corresponding drugs were used in various groups. All the mice were sacrificed 2 weeks after treatment. Sampled the blood of mice for determination of serum creatinine( Scr) and blood urea nitrogen( BUN). Renal tissues also collected to observe the pathological changes of kidney by Masson and HE staining. Results: Compared with the sham operation group,Scr and BUN level in model group increased significantly( P < 0. 001),and renal interstitial fibrosis was obvious,percentage of positive rate of collagen staining increased( P < 0. 001). The level of Scr and BUN in treatment group were obviously decreased compared with model group( P < 0. 001),and the degree of renal interstitial fibrosis in treatment group was significantly reduced,while the percentage of collagen staining decreased.( P < 0. 001). At the same time,wyxz group was better than valsartan group. Conclusion: Wenyangxiaozheng decoction can improve the renal function of UUO mice and delay the process of renal interstitial fibrosis,the effect is better than valsartan capsule.
