摘要
目的·探讨利拉鲁肽对高糖诱导人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)miR-146b-3p及白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、环氧化酶-2(cyclooxygenase-2, COX-2)表达的影响。方法·高糖组和对照组HUVECs分别以高浓度葡萄糖(25 mmol/L)及正常浓度葡萄糖(7 mmol/L)处理24 h,再以利拉鲁肽分别干预上述2组细胞。定量聚合酶链反应(quantitative polymerase chain reaction, qPCR)检测各组miR-146b-3p、IL-6、TNF-α、COX-2的表达变化。抑制miR-146b-3p后观察炎症因子及COX-2的表达变化。结果·在高糖诱导的HUVECs中,miR-146b-3p的表达下降,IL-6、TNF-α及COX-2的表达均升高。抑制mi R-146b-3p表达会显著升高高糖诱导的HUVECs中IL-6、TNF-α及COX-2的表达。利拉鲁肽可显著升高高糖诱导的HUVECs中miR-146b-3p的表达,降低IL-6、TNF-α及COX-2的表达。结论·在高糖环境下,利拉鲁肽可能通过升高miR-146b-3p的表达而降低IL-6、TNF-α及COX-2的表达,从而改善炎症和血管内皮功能。
Objective · To investigate the effects of liraglutide on glucose induced expression of miRNA-146 b-3 p, tumor necrosis factor-a(TNF-α),interleukin-6(IL-6), and cyclooxygenase-2(COX-2) in human umbilical vein endothelial cells(HUVECs). Methods · HUVECs were grown in the medium with glucose of high concentration(25 mmol/L) or normal concentration(7 mmol/L) for 24 h, then stimulated with liraglutide. Quantitative polymerase chain reaction(qPCR) was performed to detect the expression of miR-146 b-3 p, IL-6, TNF-α and COX-2. The expression of IL-6, TNF-α and COX-2 were detected after HUVECs were transfected with anti-miR-146 b-3 p. Results · The expression of miR-146 b-3 p was decreased in high glucose induced cells, while the expression of IL-6, TNF-α and COX-2 were increased. Silenced expression of miR-146 b-3 p increased the expression of IL-6,TNF-α and COX-2. Liraglutide increased the expression of miR-146 b-3 p and decreased the expression of IL-6, TNF-α and COX-2 in high glucose induced cells. Conclusion · Liraglutide may relieve inflammation and improve vascular endothelial function in hyperglycemia condition through regulating miR-146 b-3 p to decrease expression of IL-6, TNF-α and COX-2.
