摘要
为了解河南济源地区奶牛肝片吸虫感染情况,以期为该病的防制和进一步研究奠定基础,本研究采用尼龙袋集卵法对奶牛粪便中虫卵进行收集、镜检,摸清奶牛肝片吸虫病在济源市流行情况,利用PCR技术对肝片吸虫分离株线粒体nad1基因序列进行扩增与分析。结果显示,13份样品检测为阳性,阳性率为5.41%,13个分离株nad1序列长度基本一致,为488~494bp,各分离株间同源性为99.3%~100.0%;种系发育分析结果显示,所有的济源市分离株与Genbank收录的肝片吸虫分离株位于同一大分支,与大片吸虫所属分支较远。
The aims of the present study were to investigate the prevalence and molecular identification of F.hepatica from cows at Jiyuan city,Henan province.Parasite eggs from cow were collected by nylon pocket egg concentration method.The survey showed that the prevalence of F.hepatica from cow in Jiyuan city was 5.41%.In addition,the target gene(nad1)of F.hepatica was amplified by PCR to analyse the nad1 genetic variations.It was showed that the lengths of nad1 sequences were 488~494bp,and homology of the isolates was 99.3%~100.0%.The constructed phylogenetic tree revealed that the Jiyuan isolates and F.hepatica available in GenBank were clustered in the same clade.
