响应面优化提取龙牙楤木中总黄酮的工艺研究 被引量:15

摘要 以龙牙楤木芽为原料,以单因素试验为基础,分别研究乙醇浓度、提取温度、提取时间、料液比对总黄酮提取率的影响,采用Box-Bohnken Design试验设计原理进行三因素三水平试验设计和响应面分析法优化提取工艺,并对其1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除活性进行评价。结果表明,龙牙楤木总黄酮的最佳提取条件为:提取温度68.8℃、乙醇浓度69.8%、提取时间30 min,此条件下总黄酮提取率的预测值为2.21%,与实际测定值2.20%的相对误差为0.45%,且总黄酮浓度在0.1 mg/mL~1 mg/mL对DPPH自由基有一定清除活性。 Aralia elata buds as the raw material,based on the single factor experiment,the effects of ethanol concentration,extraction temperature,extraction time and liquid-to-solid ratio on extraction yield of total flavonoids were studied respectively.The three factors-three levels experiment was designed by Box-Bohnken Design centre combination theory,the optimum extraction conditions were obtained by using response surface methodology and 1,1-diphenyl-2-picrylhydrazyl(DPPH)free radical scavenging activity were evaluated.The results showed that the optimal conditions for flavonoids extraction were:extraction temperature,68.8℃;ethanol concentration,69.8%and extraction time was 30 min.Under these conditions,the experiment yield 2.20%was close to predicted yield 2.21%,relative error was 0.45%,and total flavonoids from Aralia elata had a certain scavenging activity to DPPH·between concentration 0.1 mg/mL-1 mg/mL.
