摘要 目的优选栀子提取物的分离纯化工艺参数。方法以西红花总苷转移率和色价为综合评分指标,通过单因素试验和正交试验设计优选栀子提取物的最佳分离纯化工艺参数,并进行工业化生产验证。结果最佳分离纯化工艺为:提取液以0.5 mg/mL的浓度上HPD-100A树脂吸附,依次用12 BV纯化水、2 BV的35%乙醇以1 BV/h的流速洗脱除杂,再以同样的流速用4 BV的65%乙醇洗脱即得栀子提取物。最佳工艺条件下进行中试生产验证,3批栀子提取物的西红花总苷转移率、色价和产率均值分别为86.25%、509.82和2.11%,其RSD值分别为1.56%、0.39%和2.33%。结论该栀子提取物分离纯化工艺经过工业化生产验证,稳定、可靠。 Objective To optimize the separation and purification process parameters of Gardenia jasminoides extract.Methods The transfer rate and color value of crocus sativus glycosides were used as comprehensive scoring indexes.The optimum separation and purification parameters of Gardenia jasminoides extract were optimized by single factor test and orthogonal design, and verified by industrial production. Results The optimum separation and purification process was as follows, the extract was adsorbed by HPD-100 A resin at a concentration of 0.5 mg/mL, and then eluted with 12 BV purified water, 2 BV 35% ethanol at a flow rate of 1 BV/h. The Gardenia jasminoides extract was obtained by eluting with 65% ethanol of 4 BV at the same flow rate. Under the optimal process conditions, the pilot production was verified. The average transfer rate, color value and yield of saffron from the three batches of Gardenia jasminoides extract were 86.25%, 509.82 and 2.11%, respectively, and the RSD values were 1.56%, 0.39% and 2.33%. Conclusion The separation and purification process of Gardenia jasminoides extract has been verified by industrial production and is stable and reliable.
