摘要
目的探讨肝细胞癌(HCC)组织微小RNA-155(miR-155-5p)的水平及其与临床病理特征和预后的关系。方法收集本院2013年1月至2015年12月经手术切除的114对HCC组织和癌旁组织,采用荧光实时定量PCR(QPCR)检测以上组织的miR-155-5p水平,分析miR-155-5p水平与HCC临床病理特征(性别、年龄、肿瘤数目、肿瘤大小、分化程度、肿瘤包膜、TNM分期、Child-Pugh分级和甲胎蛋白水平)的关系,根据随访资料分析不同miR-155-5p水平与HCC患者预后的关系。结果 QPCR检测结果显示HCC组织的miR-155-5p水平为3. 041±1. 393,高于癌旁组织的1. 167±0. 514(P<0. 05)。miR-155-5p水平与HCC患者的肿瘤数目、肿瘤大小、分化程度和TNM分期有关(P<0. 05),而与性别、年龄、肿瘤包膜、Child-Pugh分级和甲胎蛋白水平均无关(P>0. 05)。HCC患者全组获随访8~60个月,中位随访时间36. 0个月,中位总生存期(OS)为46. 0个月,其中miR-155-5p高表达60例的中位OS为37. 0个月,低表达54例的中位OS为57. 0个月,miR-155-5p低表达的中位OS大于高表达者(P<0. 05)。结论 HCC组织miR-155-5p水平上调参与HCC的发生发展,肿瘤负荷大、分化较差及分期较晚的miR-155-5p水平较高,而高miR-155-5p水平的预后较差,该分子可能作为一个评估HCC预后的潜在指标。
Objective To investigate the level of microRNA-155-5p( miR-155-5p) in hepatocellular carcinoma( HCC) tissues and its relationship with clinicopathological parameters and prognosis. Methods A total of 114 pairs of HCC tissues and paracancerous tissues were collected from January 2013 to December 2015. The levels of miR-155-5p in the above tissues were detected by real-time fluorescence quantitative PCR( QPCR). The relationships between miR-155-5p levels and the clinicopathological parameters of HCC( sex,age,number of tumor,tumor size,degree of differentiation,tumor capsule,TNM stage,Child-Pugh grade and alpha-fetoprotein level) were analyzed. According to the follow-up data,the relationship between different levels of miR-155-5p and prognosis of HCC patients was analyzed. Results The results of QPCR showed that the level of miR-155-5p in HCC tissues was 3. 041± 1. 393,which was higher than 1. 167±0. 514 in paracancerous tissues( P<0. 05). The level of miR-155-5p was correlated with the number of tumor,tumor size,degree of differentiation and TNM stage in HCC patients( P<0. 05),but not with sex,age,tumor capsule,ChildPugh grade and alpha-fetoprotein level( P>0. 05). HCC patients were followed up for 8 to 60 months,with a median follow-up time of36. 0 months and a median overall survival( OS) of 46. 0 months. The median OS of 60 cases with high-expression of miR-155-5p was37. 0 months,and that of 54 cases with low-expression was 57. 0 months. The median OS of low-expression of miR-155-5p was higher than that of high-expression( P<0. 05). Conclusion The up-regulation of miR-155-5p in HCC tissues is involved in the occurrence and development of HCC. The level of miR-155-5p with high tumor load,poor differentiation and late staging is higher,and the prognosis of high miR-155-5p level is worse. This molecule may be a potential prognostic indicator of HCC.
