摘要 目的:探讨LncRNA H19、miR-107和AQP1在脓毒症和脂多糖(LPS)诱导的炎症反应中的潜在机制。方法:选取收治的15例脓毒症患者为脓毒症患者组,同期健康体检者15名为健康对照组。将pc DNA3. 1/H19(pc-H19)与其阴性对照(pc-NC)转染入LPS诱导的293T细胞中。实时定量PCR检测LncRNA H19、miR-107和AQP1的表达,ELISA检测TNF-α、IL-1β和IL-6的水平。荧光素酶报告分析检测LncRNA H19、miR-107和AQP1之间的相互作用。结果:与健康对照组相比,脓毒症患者组血浆中H19和AQP1的表达降低,miR-107的表达增加,血清TNF-α、IL-6和IL-1水平增加。H19过表达增加LPS诱导的H19和AQP1的表达,降低miR-107的表达以及TNF-α、IL-6和IL-1水平。pc-H19转染可显著提高AQP1-3’UTR和miR-107共转染对荧光素酶活性的抑制。结论:H19为AQP1上miR-107的ceRNA,抑制脓毒症中的炎症反应。 Objective:To explore the underlying mechanisms of LncRNA H19,miR-107 and AQP1 on sepsis and lipopolysaccharide(LPS)-induced inflammatory response.Methods:15 patients with sepsis were selected,and 15 healthy subjects were as healthy control group.The pcDNA3.1/H19(pc-H19)and its negative control(pc-NC)were transfected into LPS-induced 293T cells.The expressions of LncRNA H19,miR-107 and AQP1 were detected by real-time quantitative PCR,and the levels of TNF-α,IL-1βand IL-6 were detected by ELISA.The interaction among LncRNA H19,miR-107 and AQP1 was analyzed by Luciferase reporter assay.Results:Compared with healthy subjects,the expressions of H19 and AQP1 in plasma of patients with sepsis were decreased,the expression of miR-107 was increased and the serum levels of TNF-α,IL-6 and IL-1 increased in sepsis patients.H19 overexpression increased LPS-induced expressions of H19 and AQP1,decreased the expression of miR-107 and the levels of TNF-α,IL-6 and IL-1.The pc-H19 transfection significantly increased the inhibition of luciferase activity induced by co-transfection of AQP1-3’UTR and miR-107.Conclusion:H19 is a ceRNA of miR-107 on AQP1,which inhibits the inflammatory response in sepsis.
