摘要
目的探讨白三烯A4水解酶(leukotriene A4 hydrolase,LTA4H)基因多态性与深圳地区汉族人群肺结核发病的关系,并结合临床指标分析,筛选功能性基因单核苷酸(single nucleotide polymorphism,SNP)。方法利用TaqMan探针技术进行基因分型,分析深圳汉族人群中958例结核病患者和945名健康对照的LTA4H基因位点rs17525495的多态性,明确基因多态性与结核病易感性的关系。结果在SNP位点rs17525495基因频率分析中,结核组CC基因型频率(11.9%)明显高于对照组(6.1%)(P<0.05),结核组G等位基因频率(32.7%)明显高于对照组(29.0%)(χ~2=6.20,P=0.0064),结核组G等位基因频率(32.7%)明显高于对照组(29.0%)(χ~2=6.20,P=0.0064)。将ESR及CT评分按照rs17525495位点的基因型进行分层分析,在三种基因型中无统计学差异。结论 LTA4H基因rs17525495位点等位基因C可能与肺结核发病相关,等位基因T可能为保护性基因。但是rs17525495位点可能不是功能性SNP。
Objective The aim of this study was to survey the relationship between polymorphism of LTA4H gene and susceptibility to tuberculosis(TB)in Han nationality,and to screen functional SNP by incorporating withclinical data.Methods The single SNP site was detected by TaqMan probe technology.Polymorphism of susceptibility gene site was analyzed between 958 patients and 945 healthy controls.The relationship between polymorphism of LTA4H gene and susceptibility to tuberculosis in Han nationality was investigated.Results After analyzing the genotypic-frequency,CC in the patient groups was significantly higher than that of control groups(P<0.05).Similarly,The C allele frequency in patient groups was significantly higher than that of control groups(χ2=6.20 P=0.0064).Conclusion The LTA4H gene polymorphism site rs17525495 may be associated with susceptibility to TB.Although rs17525495 may not be a functional SNP site,allele T may be responsible for protection.
