HPLC-ELSD法测定不同产地、不同等级蕨麻中野蔷薇苷和刺梨苷的含量 被引量:5

Content determination of rosamultin and kaji-ichigoside F1 in different regions and grades of Potentilla anserina L. by HPLC-ELSD

摘要 【目的】建立HPLC-ELSD法同时测定不同产地、不同等级蕨麻中野蔷薇苷和刺梨苷含量。【方法】本实验采用色谱柱JADE-PAK ODS(250 mm×4.6 mm,5μm,ECHWAY,China),以30%乙腈-70%水为流动相,流速设置为1.0 ml/min,进样体积为20μl,柱温为30℃;ELSD检测器漂移管温度为40℃,气体压力为350 k Pa,检测信号放大倍数为8。【结果】野蔷薇苷和刺梨苷分别在34.80~754.00μg/ml(r2=0.9981)和34.23~741.65μg/ml(r2=0.9992)范围内呈现良好的线性关系;检测限和定量限分别为10.80和23.08μg/ml、14.17和28.92μg/ml;日内和日间精密度的相对标准偏差分别为0.76%和2.36%、2.06%和6.12%;稳定性RSD均小于3.40%;加样回收率分别为97.57%和99.16%,且RSD均小于5.37%。11批不同产地和等级的蕨麻药材中,野蔷薇苷的含量在0.644(g/g)‰~1.453(g/g)‰之间,刺梨苷的含量在0.312(g/g)‰~0.563(g/g)‰之间。【结论】本实验所建立的HPLCELSD方法,可以快速、简便、准确地测定出蕨麻药材中野蔷薇苷和刺梨苷的含量。来源于青藏高原产地的蕨麻药材中野蔷薇苷和刺梨苷含量更高;相同产地的蕨麻,等级越高(颗粒越饱满),野蔷薇苷和刺梨苷的含量越低。 【Objective】To establish a method to determine the contents of rosamultin and kaji-ichigoside F1 in different regions and grades of Potentilla anserina L.by HPLC-ELSD.【Methods】HPLC-ELSD was carried out on JADE-PAK ODS(250 mm×4.6 mm,5μm,ECHWAY,China)column with isocratic elution of acetonitrile:water(30:70,v/v).The flow rate was 1.0 ml/min,and injection volume was 20μl.The temperature of the column was 30℃.Drift tube temperature of the ELSD system was set to 40℃,and the pressure of ELSD was 350 kPa.The signal of ELSD was enlarged 8 times.【Results】The regression equation revealed a good linear relationship(linear ranges=34.80~754.00μg/ml,r2=0.9981,RSD=4.85%)in rosamultin with test ranges.The regression equation also revealed a good linear relationship(linear ranges=34.23~741.65μg/ml,r2=0.9992,RSD=5.37%)in kaji-ichigoside F1 with test ranges.The detection limits and quantitation limits were 10.80 and 23.08μg/ml,14.17 and 28.92μg/ml respectively.The relative standard deviations for intra-day and inter-day precision were 0.76%and 2.36%,2.06%and 6.12%,respectively.The stability RSD was less than 3.40%.The recovery rates were 97.57%and 99.16%,respectively.And the RSD was both less than 5.37%.In 11 batches of Potentilla anserina L.with different regions and grades,the content of rosamultin was between 0.644(g/g)‰~1.453(g/g)‰,and the content of kaji-ichigoside F1 was between 0.312(g/g)‰~0.563(g/g)‰.【Conclusion】The proposed HPLC-ELSD method is fast,simple,precise for the content determination of rosamultin and kaji-ichigoside F1 in Potentilla anserina L.,Potentilla anserina L.from the Tibetan plateau contains more rosamultin and kaji-ichigoside F1.For Potentilla anserina L.from the same region,the higher grade,the lower contents of rosamultin and kaji-ichigoside F1.
