赶黄草中肝脏保护作用化学成分的分离与鉴定

Isolation and identification of hepatoprotective constituents from Penthorum chinense

【目的】研究赶黄草中肝保护的有效成分。【方法】采用不同的化学分离手段,包括硅胶柱层析以及Sephadex LH-20凝胶柱层析等方法,对赶黄草乙酸乙酯部位进行化学分离;同时结合不同的波谱解析手段对分离的化合物进行结构鉴定,采用H_2O_2诱导的HL-7702细胞进行肝保护有效成分的检测。【结果】从赶黄草乙醇提取物中分离出10个黄酮类化合物,分别为木犀草素(1)、槲皮素(2)、芹菜素(3)、乔松素-7-O-β-D-葡萄糖苷(4)、槲皮苷(5)、槲皮素-3-O-α-L-阿拉伯糖苷(6)、山奈酚-3-O-α-L-呋喃阿拉伯糖苷(7)、山奈酚-3-O-α-L-鼠李糖苷(8)、4-deoxy-phlorizin(9)、2',4',6'-trihydroydihydrochalcone-4'-O-β-Dglucoside(10);其中化合物4、7和10的OD_(450)值分别为0.317±0.017、0.287±0.017、0.412±0.017。【结论】化合物6和8为首次从扯根菜属植物中分得,化合物7、9、10为首次从虎耳草科植物中分得,其中化合物4、7和10具有较强的肝细胞保护作用。

【Objective】To study the hepatoprotective constituents of Penthorum chinense.【Methods】 Ethyl acetate fractions of Penthorum chinense were Chemically separated by different chemical separation methods, including silica gel column chromatography and Sephadex LH-20 gel column chromatography. Their structures were identified by different spectral analysis methods, and the hepatoprotective effects were evaluated through H2O2-induced HL-7702 cells.【Results】Ten flavonoid compounds were isolated from the ethanol extract of Penthorum chinense and identified as luteolin(1), qercetin(2), apigenin(3), pinocembrin-7-O-β-Dglucopyranoside(4), querctrin(5), quercetin-3-O-α-L-arabinoside(6), kaempferol-3-O-α-L-arabinoside(7), kaempferol-3-O-α-L-rhamnopyranoside(8), 4-deoxy-phlorizin(9), and 2’,4’,6’-trihydroydihydrochalcone-4’-O-β-D-glucoside(10). And OD450 values of 0.317±0.017、0.287±0.017、0.412±0.017 respectively as determining compounds 4, 5 and 10.【Conclusion】Compounds 6 and 8 were isolated from the genus Penthorum for the first time and compounds 7, 9, 10 were isolated from the family Saxifragaceae for the first time. Compounds 4, 5 and 10 show stong hepatoprotective effects.
