摘要
目的探讨Ⅱ型登革病毒DNA疫苗pVAX1-D2ME对不同性别小鼠的免疫保护效果的差异。方法利用pVAX1-D2ME质粒分别免疫雄性和雌性小鼠,每次免疫前及末次免疫后取其血清,检测体液免疫应答的各项指标;并于末次免疫后两周攻毒,观察小鼠的生存率。结果不同性别小鼠对该疫苗均产生良好的免疫应答,免疫应答具有Th1趋向性;且免疫后不同性别小鼠均可抵抗致死量病毒的攻击,生存率为100%。结论小鼠性别对免疫应答和免疫保护无明显影响,pVAX1-D2ME免疫小鼠能够诱导产生良好的免疫应答和免疫保护作用。
Objective To investigate the difference in protective efficacy of dengue serotype Ⅱ virus DNA vaccine pVAX1-D2 ME between male and female mice. Methods Male and female mice were separately immunized with pVAX1-D2 ME plasmids. Sera were obtained before each immunization and after the last immunization to evaluate the humoral immune responses. Two weeks after the last immunization, the mice were challenged with the dengue serotype Ⅱ virus(DENV2) to observe the difference of survival rate between male and female mice. Results Male and female mice showed no significant differences in the levels of specific antibodies produced by this vaccine. The immune response showed a Th1-type tendency. All the mice immunized showed complete protection against the lethal dose of virus with 100% survival rate. Conclusions The pVAX1-D2 ME could induce strong immune response and effective protection against DENV2 in mice, and these effects are not related to the gender of mice.
