细胞外信号调节激酶1/2诱导痛觉过敏机制的研究进展 被引量:3

摘要 细胞外信号调节激酶1/2(ERK1/2)是一类高度保守的丝氨酸/苏氨酸蛋白激酶,属于促分裂原活化的蛋白激酶家族。哺乳动物神经系统内ERK1/2参与外周和中枢痛觉信号的调制,在细胞内信号调节和细胞间信息交流过程中扮演重要角色。背根神经节和脊髓背角内ERK1/2能被伤害性刺激信号异常激活,活化的ERK1/2可通过转录或非转录途径促进多种类型疼痛的形成和维持,抑制其活化能显著缓解疼痛,但详细机制及两者作用的鉴别尚未完全明确。随着对疼痛研究的日益深入,ERK1/2参与疼痛进程的理论知识不断得到补充,为疼痛治疗提供了新思路。 Extracellular signal-regulated kinase 1/2(ERK1/2)is a category of serine and threonine protein kinases,which belong to mitogen-activated protein kinases family.ERK1/2 has been proved to be involved in the modulation of peripheral and central pain signals in mammalian nervous system,playing an important role in the intracellular signaling regulation and intercellular communication.ERK1/2 can be activated abnormally by noxious stimulation in dorsal root ganglion and spinal dorsal horn,which promots the formation and maintenance of different types of pain though transcriptional or non-transcriptional ways.Inhibiting the activation of ERK1/2 can significantly relieve pain.However,the detailed mecha-nisms and function identification between ERK1 and ERK2 have not been fully clarified.As the researches of pain go deeper,the theoretical knowledge of ERK1/2 participating in pain process is constantly supplemented,providing new ideas for the pain management.
