香椿子正丁醇提取物改善糖尿病肾病肾小球内皮细胞炎症及机制研究 被引量:6

Extract of Toona sinensis improves glomerular endothelial cell inflammation in DN and its potential mechanism

摘要 为研究香椿子正丁醇提取物(n-butyl alcohol extract of Toona sinensis,NBAE)对糖尿病肾病肾小球内皮细胞炎症的作用及机制,采用Wistar雄性大鼠,STZ注射造模,成功后分为DN组、DN+NBAE干预组,另设对照组。8周后取血测生化指标,取肾脏行HE和PAS染色,并行免疫组化检测MCP-1、ICAM-1、磷酸化p65的表达。以高糖(HG)、HG+NBAE、HG+NF-κB阻断剂吡咯烷二硫基甲酸盐(pyrrolidine dithiocarbamate,PDTC)刺激肾小球内皮细胞,采用Western blot法检测相关蛋白的表达。结果显示,与DN组大鼠相比,DN+NBAE组大鼠血糖明显降低,肾脏损伤减轻,相关蛋白表达均减少。细胞水平,NBAE明显降低MCP-1、ICAM-1的表达,差异具有统计学意义(P <0. 01),各指标改变情况与PDTC处理组类似。这表明NBAE明显改善DN肾小球内皮细胞的炎症,推测可能与抑制NF-κB信号通路有关。 In order to investigate the effect of n-butyl alcohol extract of Toona sinensis(NBAE)on the inflammation of glomerular endothelial cells in diabetic nephropathy(DN)and to explore the possible mechanisms,Wistar male rats were injected with STZ.After successful modeling,STZ rats were divided into DN grouPand DN+NBAE intervention group.After 8 weeks,the rats were sacrificed which blood sample were tested for BUN and Scr and the kidneys were collected for HE staining and PAS staining.MCP-1,ICAM-1 and phosphorylation of p65 were detected by immunohistochemistry staining.After stimulating glomerular endothelial cells in vitro with 25 mmol/L high glucose(HG),HG+NBAE,and HG+NF-kB p65 blocker PDTC,MCP-1,ICAM-1 and phosphorylated p65 were detected by Western blot.The results showed that compared with the DN group,the blood glucose was significantly reduced and the injury was alleviated in DN+NBAE group.Otherwise,MCP-1,ICAM-1 and phosphorylated p65 were all reduced.In vitro,NBAE significantly decreased the expression of MCP-1 and ICAM-1(P<0.01),which were similar with the treatment of NF-kBp65 blocker PDTC.In conclusion,it can be proved that endothelial inflammation of DN can be alleviated by NBAE,which is presumably related to inhibition of NF-kB signaling pathway.
