水飞蓟宾改善APP/PS1双转基因小鼠认知能力氧化应激相关机制研究 被引量:2

摘要 目的:研究水飞蓟宾对于APP/PS1双转基因小鼠学习记忆障碍的改善作用,并从氧化应激及突触角度,探究相关机制。方法:应用Morris水迷宫实验检测水飞蓟宾对APP/PS1小鼠的空间学习记忆障碍改善作用;采用ELISA法测定脑皮质组织中MDA、GSH、SOD含量;电镜观察突触的超微结构;采用ELISAGAP-43的表达。结果:水飞蓟宾能显著改善APP/PS1模型小鼠的学习记忆障碍;降低脑内MDA含量,增加SOD、GSH含量;改善突触结构损伤;增加SYP、GAP-43蛋白的表达。结论:水飞蓟宾具有改善学习记忆障碍的作用,其作用机制可能与减轻氧化应激反应,改善突触结构和功能有关。 Objective:To investigate the effect of silibinin on learning and memory impairment in APP/PS1 transgenic mice,and to explore related mechanisms from oxidative stress and synaptic structure and function.Methods:Morris water maze test was used to detect the effect of silibinin on spatial learning and memory impairment in APP/PS1 mice.ELISA was used to detect the content of MDA,GSH and SOD in the cortex tissue.Electron microscope was used to observe the ultra-structure changes of synapses and ELISA was used to detect the expression of synapse-associated proteins SYPand GAP-43 in the cortex tissue.Result:Silibinin significantly ameliorated learning and memory impairment in APP/PS1 mice,decreased the content of MDA and increased the content of GSH and SOD in the cortex tissue,improved the ultra-structure of synapses,and increased the expression levels of synaptic related protein SYPand GAP-43 in the cortex tissue.Conclusion:The cognition-improving effects of silibinin probably result from inhibition of oxidative stress and improvement of synaptic structure and function.
