非对称结构15/21小RNA dsE-661促进前列腺癌细胞DU145凋亡的效果分析 被引量:1

摘要 目的分析非对称结构15/21小RNA dsE-661促进前列腺癌细胞DU145凋亡的效果。方法以非对称结构15/21小RNA dsE-661转染前列腺癌细胞DU145(转染组,n=7),以未转染前列腺癌细胞DU145为对照(对照组,n=7)MTT法检测细胞增殖抑制率,Western blot检测转染后细胞凋亡蛋白的表达水平。结果与对照组相比,转染组细胞的增殖被明显抑制(P<0.05),并呈时间依赖性;转染组细胞的Bax、Caspase-3、Caspase-6和Caspase-9水平明显升高而Bcl-2水平明显降低,差异有统计学意义(P<0.05)。结论非对称结构15/21小RNA dsE-661能有效促进前列腺癌细胞DU145的凋亡。 Objective To analyze the effect of asymmetric structure 1521 small RNA on the apoptosis of prostate cancer cell DU 145.Methods Transfected prostatic cells DU145(transfection group,n=7)with asymmetric structure 15/21 small RNA dsE-661.Western blot was used to detect the expression level of apoptotic protein after transfection.The DU145 of untransfected prostate cells was used as control(control group,n=7).Results Compared with the control group,the proliferation rate of transfected cells was significantly inhibited(P<0.05),in a time-dependent way.In transfection group,the levels of Bax,Caspase-3,Caspase-6 and Caspase-9 increased significantly,while the level of Bcl2 decreased significantly(P<0.05).Conclusion Asymmetric structure 15/21 small RNA dsE-661 can effectively promote the apoptosis of prostate cancer cell DU145.
