摘要
本试验旨在研究FASN基因在藏猪(ZB)、大约克猪(YH)、滇南小耳猪(X)肝脏、背脂、背最长肌组织中的表达模式。试验分别选取6月龄5头生长状况良好的藏猪、大约克猪、滇南小耳猪分成3组,利用实时荧光定量PCR检测肝脏、背脂、背最长肌组织中FASN基因的表达水平。研究结果发现,在肝脏和背脂组织中ZB组和X组的FASN基因表达水平均极显著高于(P<0.01)YH组,而在ZB组和X组之间差异不显著(P>0.05)。在背最长肌组织中,ZB组的表达量极显著(P<0.01)高于YH组和X组,但YH组与X组之间差异不显著(P>0.05)。在同一品种内,背脂组织中FASN基因的表达水平均显著高于肝脏和背最长肌组织,但在不同品种内呈现模式不完全相同。在ZB组内,FASN基因表达水平从高至低依次是背脂>背最长肌>肝脏;在X组内,顺序依次是背脂>肝脏>背最长肌;在YH组内,背脂组织显著高于肝脏和背最长肌,而后二者之间差异不显著。本试验表明,在,3个不同组织器官中FASN基因在背脂中表达量最高且在藏猪和滇南小耳猪中的表达水平高于大约克猪,说明在3个组织中FASN表达模式虽有差异但其表达水平与脂肪含量都是显著正相关的。上述结果为全面解析FASN在猪背脂、肝脏和背最长肌组织的生物学功能奠定了基础。
The aim of this study is to investigate the expression pattern of FASN gene in the liver, the back fat, and the longest muscle of Tibet(ZB), Yorkshire(YH) and Diannan small ear pig(X). Three groups, ZB, YH and X, were set, and each group consisted of five health pigs. The level of FASN mRNA was investigated in liver, backfat and longissimus dorsi from the three groups. In backfat and liver tissues, high level of FASN was observed in ZB and X group compared to YH group(p<0.01), but no significant difference was found between ZB and X group(p >0.05). In longissimus dorsi tissue, high level of FASN was identified in ZB group in relative to YH and X group(p<0.01), but no difference was found between YH and X group(p >0.05). For a given breed, expression level of FASN was significantly higher in backfat tissue than liver and longissimus dorsi tissues, whereas expression pattern was various. In ZB group, mRNA level of FASN from high to low was backfat > longissimus dorsi > liver;In X group, the order was backfat > liver > longissims dorsi;In YH group, the highest level was observed in backfat tissue but it was not significantly different between longissimus dorsi and liver tissues. The results of this experiment showed that FASN gene was expressed in the back fat in three different organs and the expression level in Tibetan pigs and Diannan small pigs was higher than that in the Yorkshire, indicating that the FASN expression patterns were different in the three tissues. However, its expression level was significantly positively correlated with fat content.In summary, our result could lay the foundation for comprehensive dissection of biological function of FASN gene.
