摘要
目的探讨Ang(1-7)对室颤大鼠心肺复苏后心肌损伤及心功能的保护作用。方法建立室颤大鼠心肺复苏后模型(CA-CPR-ROSC),38只SPF级SD大鼠随机分为空白对照组(n=6)、CA-CPR-ROSC实验组(n=20)、ROSC后Ang(1-7)实验组(n=12);根据30min复苏结局分为ROSC成功组(S组,n=14)与ROSC失败组(F组,n=6),记录复苏1、2、4、6h大鼠心率、平均动脉压、±LVdp/dtmax变化,ELISA法测定血清Ang(1-7)水平;12只获得ROSC成功的大鼠分为ROSC对照组(n=6)、Ang(1-7)组(n=6),记录给予Ang(1-7)2、4、6h后心率、平均动脉压、±LVdp/dtmax变化,6h后采集全血标本,ELISA法测定血清cTnT、BNP水平。结果与对照组相比,ROSC后HR、MAP、±LVdp/dtmax下降均有明显差异(P<0.01);与F组相比,S组大鼠血清中Ang(1-7)水平明显升高(P<0.01)。与对照组比较,Ang(1-7)组大鼠的HR、MAP、±LVdp/dtmax绝对值均下降、6h后血清BNP及cTnT水平升高,差异均有统计学意义(P<0.05);与ROSC对照组比较,Ang(1-7)组大鼠在ROSC后4、6h的±LVdp/dtmax绝对值升高,在ROSC后6h血清BNP、cTnT水平降低,差异均有统计学意义(P<0.05)。结论Ang(1-7)对室颤大鼠心肺复苏后心肌损伤及心功能有保护作用。
Objective To investigate the protective effect of Ang(1-7)on myocardial injury and cardiac function in rats with ventricular fibrillation after cardiopulmonary resuscitation.Methods Establishment of a model of cardiopulmonary resuscitation(CA-CPR-ROSC).38 SPF SD rats were randomly selected from the blank control group(n=6),CA-CPR-ROSC experimental group(n=20),Ang(1-7)experimental group(n=12)after ROSC,according to the 30-minute recovery outcome,the ROSC successful group(S group,n=14)and the ROSC failure group(F group,n=6).The heart rate,mean arterial pressure and±LVdp/dtmax of the rats at 1h,2h,4h and 6h were recorded.The serum Ang(1-7)level was determined by ELISA.The 12 rats with successful ROSC were divided into ROSC control group,Ang(1-7)group,recorded heart rate,mean arterial pressure,±LVdp/dtmax change after 2 hours,4h,6h of Ang(1-7)administration,collected after 6 hours whole blood samples were taken and serum cTnT and BNP levels were determined by ELISA.Results Compared with control group which HR,MAP,±LVdp/dtmax decreased significantly after ROSC(P<0.01).Compared with F group,serum Ang(1-7)level in S group significantly increased,there were statistical significantly differences(P<0.01).Compared with control group,the absolute values of HR,MAP and±LVdp/dtmax in Ang(1-7)group decreased,and serum BNP and cTnT levels increased after 6h,there were statistical significantly differences(P<0.05).Compared with the ROSC control group,the absolute value of±LVdp/dtmax increased in the Ang(1-7)group at 4h and 6h after ROSC,and the serum BNP and cTnT levels decreased at 6h after ROSC,there were statistical significantly differences(P<0.05).Conclusion Ang(1-7)has protective effects on myocardial injury and cardiac function in rats with ventricular fibrillation after cardiopulmonary resuscitation.
