miR-451在缺血性脑卒中表达的相关性研究

Correlation between miR-451 expression in peripheral blood of patients with acute stroke

目的探讨不同时间点miR-451在缺血性脑卒中患者外周血外泌体中表达量的改变,探索其成为脑卒中生物标记物的可行性。方法收集大连大学附属新华医院神经内科30例缺血性脑卒中患者发病后第1天和第7天血液标本,以及16例非脑卒中患者的血液标本作为对照组。分离血清中外泌体并采用实时荧光定量PCR技术检测患者血清外泌体中miR-451的表达量。通过ROC曲线分析血清miR-451水平对脑卒中的诊断效能。结果相较于对照组,脑卒中患者发病后第1天和第7天血清外泌体中miR-451的表达均有显著增高。此外,ROC曲线分析结果显示,1d组和7d组miR-451曲线下面积(AUC)均为1;取相对表达值1为截断点,血清miR-451预测脑卒中患者发病的灵敏度为100%,特异度为100%。结论缺血性脑卒中患者外周血外泌体中miR-451的表达与缺血性脑卒中的发生有紧密的相关性,可作为一种临床诊断的潜在生物指标。

Objective To investigate the changes of miR-451 expression in peripheral blood exosomes of patients with ischemic stroke at different time points,and to explore its feasibility as a biomarker for stroke.Methods The blood samples from the first and seventh day after onset of 30 patients with ischemic stroke in the Department of Neurology of our hospital and 16 non stroke patients were collected as control group.Serum exosomes were separated and the expression of miR-451 in serum exosomes was detected by real time fluorescent quantitative PCR.The ROC curve was used to analyze the diagnostic efficacy of serum miR-451 level in stroke.RESULTS Compared with the control group,the expression of miR-451 in serum exosomes on the first and seventh day after onset of stroke patients was significantly increased.In addition,ROC curve analysis showed that the area under the miR-451 curve(AUC)was 1 in the 1 day group and the 7 day group;the relative expression value 1 was the cut off point,and the sensitivity of serum miR-451 to predict the onset of stroke patients was 100%.The specificity is 100%.Conclusion The expression of miR-451 in peripheral blood exosomes of patients with ischemic stroke is closely related to the occurrence of ischemic stroke,and it can be used as a potential biomarker for clinical diagnosis.
