摘要
目的探索胶质瘤中的干细胞的成球能力与治疗效果的关系。方法选取41例外科手术切除的胶质瘤样本,术后均接受放疗及TMZ化疗。将胶质瘤组织进行肿瘤干细胞富集成球培养,并分析肿瘤细胞成球率及肿瘤球对于1~60Gy剂量放疗和3.9μmol/L~1mmol/L TMZ化疗的敏感性与患者生存率的相关性。结果胶质瘤细胞成球能力与患者较差的总生存期或无进展生存期(progression-free survival,PFS)具有相关性。胶质瘤细胞成球后对于放化疗有较强的抗性,放射半致死计量>12Gy的肿瘤球来源的患者具有较短的PFS,生存期也较短。肿瘤球对于TMZ化疗的敏感性和患者的生存率有直接联系,TMZ的半数抑制剂量<50μmol/L的肿瘤球来源的患者具有更长的OS和PFS。结论肿瘤球放疗敏感性检测可以指导临床对患者治疗的策略选择,以期达到更好的治疗效果。
Objective To explore the relationship between the ability of stem cells in gliomas and therapeutic effects.Methods Forty one surgically removed glioma samples were selected and received postoperative radiotherapy and TMZ chemotherapy.The glioma tissues were subjected to tumor stem cell rich bulb culture,and the correlation between tumor cell glomeration rate and tumor sphere sensitivity to 1 to 60 Gy dose radiotherapy and 3.9μmol/L to 1 mmol/L TMZ chemotherapy was analyzed.Results The ability of glioma cells to become globular was associated with poor overall survival or progression-free survival(PFS).Glioma cells have strong resistance to radiotherapy and chemotherapy after pelleting.Patients with tumor-derived radiation>40 Gy have a shorter PFS and shorter survival.The sensitivity of the tumor sphere to TMZ chemotherapy is directly related to the patient's survival rate.The tumor-derived patients with TMZ's half inhibitor<50μmol/L have longer OS and PFS.Conclusion The sensitivity test of tumor ball radiotherapy can guide the clinical choice of patients for treatment,in order to achieve better therapeutic effect.
