摘要
目的:探讨膜联蛋白A5低表达对人胃癌细胞Galectin-3表达的影响。方法:人胃癌HGC-27细胞和MGC-803细胞分别分为siRNA干扰组、阴性对照组和空白对照组,siRNA干扰组、阴性对照组细胞分别转染靶向膜联蛋白A5的si RNA Oligo和阴性对照siRNA Oligo。采用Western blotting法和qRT-PCR法检测HGC-27细胞和MGC-803细胞膜联蛋白A5、Galectin-3蛋白和m RNA的表达情况。结果:膜联蛋白A5的表达受抑制后,HGC-27细胞Galectin-3蛋白和m RNA的表达均明显降低,与阴性对照组和空白对照组相比差异具有统计学意义(P<0.05)。与阴性对照组和空白对照组相比,si RNA干扰组MGC-803细胞Galectin-3的表达无明显改变(P>0.05)。结论:膜联蛋白A5对胃癌细胞Galectin-3的表达具有调控作用,该调控作用可能与胃癌细胞的分化程度有关。
Objective:To investigate the effects of low expression of annexin A5(ANXA5)on Galectin-3 expression in human gastric cancer cells.Methods:Human gastric cancer HGC-27 cells and MGC-803 cells were grouped in siRNAinterference group,negative control group and blank control group.The cells in siRNA interference group and negative control group were respectively transfected with siRNA Oligo targeting ANXA5 and negative control siRNA Oligo.Western blotting and qRT-PCR were used to detect the ANXA5,Galectin-3 protein and mRNA expression in HGC-27 cells and MGC-803 cells.Results:After inhibiting ANXA5 expression,the Galectin-3 protein and mRNA expression in HGC-27 cells were all significantly decreased compared with HGC-27 cells in negative control group and blank control group(P<0.05).Compared with negative control group and blank control group,there had no significant changes of Galectin-3 expression in MGC-803 cells in siRNAinterference group(P>0.05).Conclusions:ANXA5 has regulatory effects on Galectin-3 expression in human gastric cells,which may be related to differentiation degree of gastric cancer cells.
