中风1号对急性期脑出血大鼠Bcl-2、Bax蛋白表达的影响

Effect of Zhongfeng No.1 Decoction on Bcl-2 and Bax Protein Expression in Rats with Acute Cerebral Hemorrhage

目的通过应用活血化瘀中药制剂中风1号治疗急性期脑出血大鼠,观察脑组织中Bcl-2和Bax蛋白表达的变化,探讨其作用机制。方法以大鼠自体尾静脉取血制作脑出血大鼠模型,造模3 d后断头取脑,应用免疫组织化学法检测脑组织中Bcl-2、Bax免疫反应阳性细胞计数。结果造模后经治疗3 d,与模型组相比,中风1号组及甘露醇组Bcl-2阳性细胞数明显升高(P <0.05),Bax阳性细胞数明显降低(P <0.05),中风1号组与甘露醇组Bcl-2/Bax比值远大于1。结论活血化瘀制剂中风1号通过调控抗凋亡基因Bcl-2、Bax的表达,对急性期脑出血起到治疗作用。

Objective:To observe the changes of Bcl-2 and Bax protein expression in brain tissue of rats with acute cerebral hemorrhage treated by Zhongfeng No.1 Decoction,and to explore its mechanism.Methods:Rat model of cerebral hemorrhage was Established by taking blood from autologous caudal vein of rats.After 3 days of modeling,the brain was removed.Bcl-2 and Bax immunoreactive positive cells were detected in brain tissue with immunohistochemical method.Results:After 3 days′treatment,the number of Bcl-2 positive cells in Zhongfeng No.1 Decoction group and mannitol group was significantly higher than that in the model group(P<0.05);the number of Bax positive cells was significantly lower(P<0.05).The ratio of Bcl-2/Bax in Zhongfeng No.1 Decoction group and mannitol group was much higher than 1.Conclusion:Zhongfeng No.1 Decoction can regulate the expression of anti-apoptotic genes Bcl-2 and Bax,playing a therapeutic role.
