摘要
目的探讨表皮生长因子协同体(EGF-c)预处理对大鼠脑缺血再灌注(IR)的保护作用及其相关机制。方法将30只雄性SD大鼠按随机数字表法分为假手术组(sham组)、缺血再灌注组(IR组)及EGF-c预处理+IR组(EGF-c组)。采用线栓法制作大鼠大脑中动脉栓塞模型(MCAO)。术后24 h,应用2%氯代三苯基四氮唑(TTC)染色法检测脑梗死体积,酶联免疫吸附测定(ELISA)法检测脑组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,蛋白免疫印迹法(Western blot)检测高迁移率族蛋白1(HMGB1)的表达。结果与sham组比较,IR组脑梗死体积增大(P <0.01),SOD活性降低(P <0.01),MDA和HMGB1的含量增加(P <0.01)。与IR组比较,EGF-c组脑梗死体积减小(P <0.05),SOD活性上升(P <0.01),MDA含量下降(P <0.01),HMGB1的表达量下降(P <0.05)。结论 EGF-c预处理可能通过改善脑组织SOD活性、降低MDA含量、下调HMGB1的表达从而对大鼠IR起到保护作用。
Objective To study the effect of epidermal growth factor-coordination(EGF-c)on cerebral ischemia reperfusion(IR)in rats as well as its mechanism.Methods Thirty male SD rats were divided into sham group,ischemia reperfusion group(IR group),EGF-c pretreatment and IR group(EGF-c group)by random number table method.The middle cerebral artery occlusion(MCAO)model of rats was made by suture method.24 hours after operation,the cerebral infarction volume was detected by 2%triphenyl tetrazolium chloride(TTC)staining,the activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in brain tissue were measured by enzyme-linked immunosorbent assay(ELISA),the expression of high mobility group protein 1(HMGB1)was examined by Western blot.Results Compared with sham group,the infarction volume was increased(P﹤0.01),the SOD activity was decreased(P﹤0.01),the contents of MDA and HMGB1 were increased in IR group(P﹤0.01).Compared with IR group,the infarction volume was decreased(P﹤0.05),SOD activity was increased(P﹤0.01),the content of MDA was decreased(P﹤0.01),the expression of HMGB1 was decreased in EGF-c group(P﹤0.05).Conclusion EGF-c pretreatment may play a protective effect on IR of rats by improving the SOD activity,decreasing the content of MDA and the expression of HMGB1.
