生脉散对2型糖尿病大鼠胰岛β细胞NF-κB/IκB及VEGF表达的影响 被引量:9

Effect of Shengmai power on VEGF expression of pancreatic β cells in type 2 diabetic rats

摘要 目的观察生脉散对2型糖尿病大鼠胰岛β细胞NF-κB/IκB及血管内皮生长因子(VEGF)表达的影响。方法将120只SD大鼠随机分为A组20只和造模组100只。A组予以基础饲料喂养。造模组基础饲料适应性喂养1周,给予高脂饲养喂养8周,一次性腹腔注射链脲佐菌素30 mg/kg建立2型糖尿病动物模型,共建模成功70只。随机将成模大鼠分为B、C、D、E、F组,每组14只。B组给予等量生理盐水灌胃,C组给予0. 2 g/(kg·d)阿司匹林灌胃,D、E、F组给予4. 7、9. 4、18. 8 g/(kg·d)生脉散汤剂灌胃,持续治疗4周。实验期间,A、B、C、D、E、F组分别死亡6、3、1、2、1、3只。检测各组治疗前后空腹血糖(FPG)、空腹胰岛素(FINS)、胰岛素敏感指数(ISI)及胰岛素抵抗(HOMA-IR),采用ELISA法检测血清VEGF,采用HE染色法观察血管壁结构改变,采用real-time PCR法检测胰岛β细胞NF-κB、IκB mRNA表达。结果治疗4周,C、D、E、F组血管内侧壁损伤明显减轻,有修复趋势,其中E、F组修复作用更明显(P <0. 05)。与B组比较,C、D、E、F组FPG、FINS、HOMA-IR显著降低,ISI显著升高(P均<0. 05);与C组比较,D组FPG、FINS升高,E、F组FPG、FINS、HOMA-IR降低,ISI升高(P均<0. 05)。与B组比较,C、D、E、F组NF-κB mRNA表达及血清VEGF水平显著降低,C、D、E、F组IκB mRNA表达显著升高(P均<0. 05)。结论生脉散能明显改善2型糖尿病大鼠胰岛素抵抗,降低血清VEGF水平,对血管壁结构有一定修复作用;其机制可能与抑制胰岛β细胞NF-κB/IκB表达有关。 Objective To observe the regulating effect of Shengmai powder on the expression levels of nuclear factor-κB/inhibitor nuclear factor-κB(NF-κB/IκB)and vascular endothelial growth factor(VEGF)in the pancreaticβcells of rats with type 2 diabetes mellitus(T2DM).Methods Totally 120 SD male rats were randomly divided into the groups A,B,C,D,E,and F.Rats in the group A were given basic diet,while rats in the other groups(B,C,D,E and F)were fed with high-fat diet combined with intraperitoneal injection of low-dose streptozotocin(STZ)to establish the rat models of diabetic obesity.After 8 weeks,the successfully established rat models were selected from each group.Rats in the group B were given the same amount of normal saline by gavage,group C 0.2 g/(kg·d)aspirin,and groups D,E and F were given 4.7,9.4 and 18.8 g/(kg·d)Shengmai powder.All treatments lasted for 4 weeks.Fasting blood glucose(FBG),fasting insulin(FINS),insulin sensitivity index(ISI)and insulin resistance(HOMA-IR)were measured before and after the above intervention.In addition,serum VEGF was detected by enzyme-linked immunosorbent assay(ELASA),the change of vascular wall structure was observed by HE staining,and NF-κB and IκB mRNA in isletβcells of rats were detected by real-time PCR.Results After 4 weeks of treatment,HE staining showed that the change of vascular wall structure was ameliorated and accompanied with tendency of repair in the groups D,E and F,especially in the latter two groups(P<0.05).Compared with the group B,the levels of FBG,FINS,and HOMA-IR decreased in the groups D,E and F,while the level of ISI increased.Compared with the group C,the levels of FBG and FINS were higher in the group D,the levels of FBG,FINS,and HOMA-IR were lower,and the level of ISI was higher in the groups E and F,with statistically significant difference(all P<0.05).Compared with the group B,the levels of NF-κB mRNA and serum VEGF decreased significantly in the groups C,D,E,and F,and the levels of IκB mRNA in the groups C,D,E and F increased significantly(P<0.05).Conclusion Shengmai powder can significantly decrease the level of insulin resistance and VEGF in serum by inhibiting the serum NF-κB/IκB expression.
