摘要
使用免疫亲和层析净化-超高效液相色谱法同时测定了生鲜牦牛乳中的黄曲霉毒素M_1(Aflatoxin M_1,AFM_1)及黄曲霉毒素M_2(Aflatoxin M_2,AFM_2)。黄曲霉毒素M_1仪器线性范围0.400-100μg/mL,仪器定量限0.400ng/mL,仪器检出限0.100ng/mL,方法检出限0.005μg/kg,回收率85.2-95.5%;黄曲霉毒素M_2仪器线性范围0.100-100μg/mL,仪器定量限0.100ng/mL,仪器检出限0.03ng/mL,方法检出限0.0015μg/kg,回收率82.1-93.8%。使用该方法对来自若尔盖地区的10个生鲜牦牛乳进行了测定,结果表明生鲜牦牛乳中黄曲霉毒素M_1和M_2安全可控。
Aflatoxin M1(AFM1)and aflatoxin M2(AFM2)in raw yak milk was determined simultaneously by an ultra high performance liquid chromatography fluorescence detection method after immunoaffinity column purification.The instrumental linear range of AFM1 is 0.40-100μg/mL and the instrumental limits of quantification and determination are 0.40ng/mL and 0.10ng/mL,respectively.The limit of determination of method is 0.005μg/kg and the average recoveries were 85.2-95.5%for AFM1.The instrumental linear range of AFM2 is 0.10-100μg/mL and the instrumental limits of quantification and determination are 0.10ng/mL and 0.03ng/mL,respectively.The limit of determination of method is 0.0015μg/kg and the average recoveries were 82.1-93.8%for AFM2.Ten raw yak milk collected from Ruoergai area were tested.The results showed that the raw yak milks were safe about AFM1 and AFM2 contaminations.
