摘要
当前,微生物细胞工厂的构建逐渐成为工业上生产大宗和精细化学品的重要手段。然而,微生物遗传背景的复杂性及表型间相互影响的不可预测性限制了人们对微生物的理性改造。因此,在缺乏遗传背景基础上仍能获得目标表型的微生物实验室进化手段逐渐成为当今生物化工领域的研究热点。近年来,随着微生物连续培养技术及体内连续进化策略的深入研究,微生物实验室进化取得了较大进展。本文中,笔者将对近年来微生物实验室进化策略进行综述,为其后续研究及应用提供借鉴。
Nowadays,microbial cell factory has been an important means of industrial production for many bulk and fine chemicals.However,the complexity of the genetic background and the unpredictability of the phenotypic interaction limit the rational design of microorganisms.Therefore,microbial laboratory evolution,that can acquire desired traits without prior genetic background,has become an important research interest in the field of biological chemistry.Recently,great achievements in continuous microbe culturing and in vivo continuous directed evolution facilitated the development of microbial laboratory evolution.In this review,recent achievements on the microbial laboratory evolution are introduced and discussed.
