Expression and significance of long non-coding RNA LINC00308 in prostate cancer

摘要 目的本研究通过分析前列腺癌(prostate cancer,Pca)患者癌及癌旁组织中差异表达的lncRNA,筛选出与Pca发生发展密切相关的lncRNA,结合lncRNA在癌和癌旁组织中的表达水平探讨其意义,为Pca的诊断和预后提供新的生物标志物和治疗靶点。方法利用TCGA数据库获取499例Pca和52例癌旁组织的基因数据,利用Perl提取lncRNA的矩阵,通过对差异表达的lncRNA进行生存分析,筛选出具有显著意义的lncRNA;通过对相关lncRNA在癌与癌旁组织中的表达水平的分析,比较其表达水平的差异性;通过生物信息学分析得到可能存在的lncRNA与靶基因的调控关系,初步阐明lncRNA对Pca患者的诊断和预后的意义。结果通过生物信息学分析筛选出具有显著性意义的差异表达的LINC00308,经过分析LINC00308在Pca的表达显著高于癌旁组织(P<0.001),且与PCA3表达水平具有相关性(Spearman系数=0.11,P=0.0096)。结论在Pca中LINC00308表达水平显著升高,其可能与Pca的发生发展密切相关,可能成为新的肿瘤标志物。 Objective To analyze the differentially expression of lncRNA in cancer tissues and adjacent normal tissues to cancer of prostate cancer(Pca),and investigate the lncRNA which was related to the development of Pca.The study could find new biomarkers and theapeutic targets for the diagnosis and prognosis of prostate cancer.Methods 499 cases of Pca and 52 cases of adjacent normal tissues to cancer were selected from TCGA database,and the survival analysis were performed to screen out the significant differentially expressed lncRNAs.The expression level of the differentially expressed lncRNA in cancer and normal tissues were compared,and the target genes to expound the regulation of lncRNAs were predicted on prostate cancer.Results The expression of LINCRNA00308 in Pca was significantly higher than that in adjacent tissues(P<0.001)and it has a correlation with the expression level of PCA3(Spearman coefficient=0.11,P=0.001).Conclusion The expression level of LINCRNA00308 was significantly increased in Pca,which might be related to the occurrence and development of Pca,and could be selected as a new biomarker.
