雌激素通过抑制Caveolin-1表达促进宫颈癌细胞生长

摘要 目的宫颈癌是女性最常见的恶性肿瘤之一,而雌激素与宫颈癌的发病有密切关系。我们选择与癌症发病关系密切的窖蛋白Caveolin-1作为研究的切入点,旨在阐明Caveolin-1在雌激素诱发宫颈癌中所发挥的作用。方法用17β-雌二醇处理Hela细胞24h、48h,免疫印迹法检测Caveolin-1表达。用siRNA沉默Hela细胞Caveolin-1基因,免疫印迹法检测Caveolin-1表达与AKT磷酸化水平改变,并用CCK8法检测Hela细胞增殖能力。结果与对照组比较,17β-雌二醇处理细胞48h后,Hela细胞Caveolin-1的表达水平显著降低(P<0.05)。Caveolin-1基因沉默后,与阴性对照组比较,p-AKT/AKT比值升高(P<0.05)。在细胞增殖实验中,与阴性对照组比较,Caveolin-1基因沉默组细胞在第72h,细胞活力明显升高(P<0.05)。结论雌激素能够通过抑制Caveolin-1表达,继而活化PI3K/AKT途径,促进宫颈癌细胞的生长。 Objective Cervical cancer is one of the most common malignant tumors in women,and estrogen is closely related to the pathogenesis of cervical cancer.We selected the caveolin-1 as the target,which is closely related to the pathogenesis of cancer.We aimed to study the role of caveolin-1 in estrogen-induced cervical cancer.Methods Hela cells were treated with 17β-estradiol for 24h and 48h,and the expression of caveolin-1 was detected by immunoblotting.The caveolin-1 gene of Hela cells was silenced by siRNA.The expression of caveolin-1 and the level of AKT phosphorylation were detected by immunoblotting.The proliferation of Hela cells was detected by CCK8 assays.Results Compared with control group,the expression of caveolin-1 in Hela cells was significantly decreased after treatment with 17β-estradiol for 48h(P<0.05).After caveolin-1 gene was silenced,the ratio of p-AKT/AKT was increased compared with negative control group(P<0.05).In the cell proliferation experiment,compared with negative control group,the cell viability of caveolin-1 gene silencing group was significantly increased at 72h(P<0.05).Conclusion Estrogen can promote the growth of cervical cancer cells by inhibiting the expression of caveolin-1 and activating PI3K/AKT pathway further.
