桑黄基因组DNA提取方法优化及分子鉴定 被引量:1

摘要 本研究以桑黄为材料,筛选出一种快速直接提取子实体DNA的方法。利用CTAB法、试剂盒及硅珠DNA纯化技术分别对桑黄子实体基因组DNA进行提取,结果表明,硅珠DNA纯化技术的提取效果最好,试剂盒方法次之。经过PCR扩增和测序得到桑黄的rDNA ITS序列,通过BLAST比对及系统进化树构建,发现所检测的样品为哈尔蒂木层孔菌Phellinus hartigii。 Using Phellinus igniariusas as research materials,a method for rapidly extracting DNA from Phellinus igniarius was screened.The genomic DNA of Phellinus igniarius was respectively extracted by CTAB method,DNA extract kit and silica bead DNA purification technology.The results showed that the extraction method of silica bead DNA purification technology was the best,and the kit method was the second.The ITS gene sequence of Phellinus igniarius was obtained by PCR amplification and sequencing.By the BlAST sequence alignment and the construction of phylogenetic tree,we found the detected sample was Phellinus hartigii.
