以甘油为碳源合成聚羟基丁酸细菌的筛选与鉴定

摘要 为降低聚羟基丁酸的生物合成成本,从武汉科技大学沁湖边采集的土壤中分离得到一株以甘油为碳源生长的菌株PHB-XYH,根据苏丹黑染色结果,可以初步推断该菌可以在胞内积累聚羟基丁酸。经形态学、生理生化特征及16S rDNA序列分析,显示菌株PHB-XYH归属于蕈状芽胞杆菌。红外光谱和气相色谱检测结果显示以甘油为碳源培养的细菌胞内提取的白色薄膜物质中含有聚羟基丁酸。而且以甘油为碳源合成的聚羟基丁酸的质量可达0. 45 g/L,含量33. 21%,提取率达到44. 42%。 In order to reduce the production cost of polyhydroxybutyrates(PHB)by microbial fermentation,strain PHB-XYH grown with glycerol as carbon source was isolated from the soil sample from the neighborhood of Qinhu in Wuhan University of Science and Technology.This bacterium was able to accumulate intracellular PHB based on Sudan Black B staining result.Strain PHB-XYH was identified as Bacillus mycoides according to its morphological,physiological and biochemical characteristics and 16S rDNA sequence analysis.The infrared spectra and gas chromatography analysis showed that the white film material extracted from strain PHB-XYH cultured with glycerol as carbon source contained PHB.The mass of PHB extracted from strain PHB-XYH cultured with glycerol was 0.45 g/L,and the content and extraction rate of PHB reached 33.21% and 44.42%,respectively.
