管花肉苁蓉药渣中多糖含量测定及抗氧化活性研究

Studies on determination of polysaccharides and its antioxidant activity in the residue of Cistanche tubulosa

目的测定管花肉苁蓉药渣中多糖含量及其抗氧化活性。方法采用水提醇沉法提取管花肉苁蓉药渣中多糖成分,采用蒽酮硫酸法测定含量,利用二苯代苦味酰基(DPPH)、羟自由基(·OH)以及铜离子还原能力清除实验综合评价管花肉苁蓉药渣中多糖的体外抗氧化活性,通过红外光谱对多糖进行初步结构鉴定。结果管花肉苁蓉药渣中多糖含量为0.025mg/g,当多糖浓度为0.04mg/mL时,对DPPH自由基清除率为59%,Vc的清除率为89%,当多糖浓度为0.03μg/mL时,对铜离子还原能力为64%,Vc的还原能力为88%,当多糖浓度为0.015mg/mL时,多糖对羟自由基清除率为70%,Vc的清除率为70%。红外光谱分析表明管花肉苁蓉药渣中多糖提取物中含有α-多糖。结论管花肉苁蓉药渣中多糖含量较高,具有抗氧化活性。

Objective To determine the content of polysaccharide and its antioxidant activity in the residue of Cistanche tubulosa deserticola.Methods Polysaccharides from Cistanche tubulosa deserticola dregs by water extraction and alcohol precipitation were extracted,and content of polysaccharides was determined by anthrone sulfuric acid method.The antioxidant activity of polysaccharides from the residue of Cistanche tubulosa deserticola was comprehensively evaluated by diphenyl acyl(DPPH),hydroxyl radical(OH)and the scavenging ability of copper ions in vitro.The preliminary structure of polysaccharides was identified by IR spectra.Results The content of polysaccharides in the residue of Cistanche tubulosa was 0.025 mg/g.When the concentration of polysaccharides was 0.04 mg/mL,the scavenging rate of DPPH free radical was 59%,and the scavenging rate of Vc was 89μg/mL.When the concentration of polysaccharides was 0.03μg/mL,and the reduction ability to copper ion was 88%.When the concentration of polysaccharides was 0.015 mg/mL,the clearance rate of polysaccharide to hydroxyl radical was 70%,and the clearance rate of Vc was 70%.Infrared spectrum analysis showed that the polysaccharides extract of Cistanche tubulosa containedα-polysaccharide.Conclusion The content of polysaccharides in the residue of Cistanche tubulosa is higher and has antioxidant activity.
