白酒大曲中一株散囊菌的鉴定及培养条件优化

摘要 以分离自扳倒井中高温大曲中的一株散囊菌(Eurotium)CICC 41584为研究对象,经菌落形态观察、理化特征试验以及分子生物学研究对菌株CICC 41584进行鉴定。在基础生长培养基上,通过单因素试验、Plackett-Burman及Box-Behnken试验对菌株CICC41584培养条件进行优化。结果表明,菌株CICC 41584被鉴定为谢瓦散囊菌(Eurotium chevalieri),该菌株最佳培养条件为蔗糖添加量40 g/L,酵母浸粉添加量27.2 g/L,硫酸镁添加量2.6 g/L,培养温度28℃,培养时间10 d,转速120 r/min,接种量2%,装液量50 m L/250 m L。在此优化条件下,菌丝干质量达到1.95 g/100 m L,较优化前提高了8.29倍。 Using Eurotium CICC 41584 isolated from Bandaojing high temperature Daqu as research objects,after colonial morphology observation,physiological and biochemical characteristics experiments and molecular biology,the strain CICC 41584 was identified.On the basic growth medium,the culture conditions of strain CICC 41584 were optimized by single factors,Plackett-Burman,and Box-Behnken experiments.The results indicated that the strain CICC 41584 was identified as Eurotium chevalieri.The optimum culture conditions of the strain were as follows:sucrose 40 g/L,yeast powder 27.2 g/L,MgSO4 2.6 g/L,culture temperature 28℃,culture time 10 d,rotate speed 120 r/min,inoculum 2%,and loaded volume 50 ml/250 ml.Under the optimal conditions,mycelium dry mass reached 1.95 g/100 ml,which was 8.29 times higher than that of before optimization.
