摘要
目的分析miR-181a和CA125在Ⅰ型、Ⅱ型子宫内膜癌患者中的表达水平以及相关性,为进一步探索子宫内膜癌的发病机制及预后提供理论依据。方法对2014年6月~2016年12月收集的81例子宫内膜组织作为实验标本,包括35例Ⅰ型子宫内膜癌、11例Ⅱ型内膜癌和20例内膜增生症以及15例正常标本。采用实时荧光定量PCR技术检验上述实验标本中miR-181a、CA125的表达水平,同时采用Spearman相关分析法分析子宫内膜癌病变中miR-181a、CA125表达的相关性。结果Ⅰ型、Ⅱ型子宫内膜癌组织的CA125、miR-181a表达均显著高于正常组织和子宫内膜增生组织(P<0.05);且Ⅱ型内膜癌组织miR-181a表达水平明显高于Ⅰ型,但其CA125表达水平显著低于Ⅰ型内膜癌组织(P<0.05);Spearman相关分析发现,在子宫内膜癌变过程中,miR-181a与CA125的表达情况呈显著负相关(r=0.816,p≤0.001)。结论 miR-181a在子宫内膜癌中呈显著高表达,其表达水平与CA125表达水平呈显著负相关,提示miR-181a有可能经过对CA125表达的调控作用来影响子宫内膜癌的发生、发展,可将其用于子宫内膜癌预后的评估。
Objective To analyze the expression levels of miR-181a and CA125 and possible correlation between them in patients with type I and II endometrial carcinoma, so as to provide theoretical basis for further exploration of pathogenesis and prognosis of endometrial carcinoma. Methods From June 2014 to December 2016, 81 endometrial tissues were collected as experimental specimens, including 35 type I endometrial carcinoma, 11 type II endometrial carcinoma, 20 endometrial hyperplasia and 15 normal tissues. The expression levels of miR-181a and CA125 in the tissues were examined by real-time fluorescence quantitative PCR, and Spearman’s correlation analysis was used to investigate the possible correlation between the miR-181a and CA125 levels in the endometrial carcinoma tissues. Results The expression levels of CA125 and miR-181a in the type I and II endometrial carcinoma were significantly higher than those in the endometrial hyperplasia and normal tissues (all p < 0.05). The expression level of miR-181a in type II endometrial carcinoma was significantly higher, while the expression level of CA125 was significantly lower, than that in type I endometrial carcinoma (all p < 0.05). Spearman’s correlation analysis showed that the expression level of miR-181a was negatively correlated with the expression level of CA125 in the carcinogenesis of endometrial carcinoma (r=-816,p < 0.000). Conclusion miR-181a is significantly overexpressed in endometrial carcinoma, and its expression level is negatively correlated with CA125, which suggestes that miR-181a may affect the occurrence and development of endometrial carcinoma by regulating the expression of CA125 in endometrial tissue, and can be used for the prognosis evaluation of endometrial carcinoma.
