摘要
目的:体外培养人树突状细胞(DC)和细胞因子诱导的杀伤细胞(CIK),探讨HPV m E7蛋白对DC和CIK细胞的影响。方法:分离培养健康人外周血中的DC和CIK细胞,加纯化的HPV m E7蛋白作为刺激因子,流式细胞仪检测DC和CIK细胞表面特异性因子。结果:成功分离培养出人外周血中DC和CIK细胞,HPV m E7蛋白刺激后细胞表面的特异性因子表达没有发生明显改变。结论:成功建立DC和CIK细胞分离培养技术,纯化后的HPV m E7可做为特异性抗原应用于宫颈癌免疫治疗研究。
Objective: Human DC and CIK cells were cultured in vitro and determine the influence of HPV mE7 protein on DC and CIK cells. Methods: DC and CIK were separated and cultured in vitro from peripheral blood of healthy people.The surface characteristics of the cells were detected before and after being stimulated by HPV mE7 protein. Results: DC and CIK cells from human peripheral blood were isolated and cultured successfully.The expression of specific factors on the surface of DC and CIK stimulated by HPV mE7 protein did not change significantly. Conclusion: DC and CIK cells were isolated and cultured successfully,and purified HPV mE7 could be used as a specific antigen for immunotherapy of cervical cancer.
