摘要
目的观察哮喘宁颗粒对哮喘模型大鼠白三烯B4(LTB4)、白三烯C4(LTC4)、C-反应蛋白(CRP表达的调节作用,探讨哮喘宁颗粒对哮喘大鼠气道相关炎症介质的影响,为哮喘宁颗粒防治哮喘提供实验依据。方法雄性Sprague-Dawley大鼠60只,随机分成5组,分别是空白对照组、模型组、哮喘宁颗粒组、布地奈德组、地塞米松组,每组12只。采用卵白蛋白联合氢氧化铝腹腔注射致敏和卵白蛋白滴鼻激发方法建立哮喘模型大鼠,采用双抗夹心酶联免疫吸附试验法检测各组大鼠肺组织匀浆中LTB4、LTC4的含量变化,速率散射比浊法检测CRP的表达变化。结果模型组大鼠肺组织中LTB4、LTC4、CRP含量均升高,与空白对照组相比差异有统计学意义(P <0. 05)。治疗后,哮喘宁颗粒组、布地奈德组、地塞米松组大鼠肺组织中LTB4、LTC4、CRP含量均降低,与模型组相比差异有统计学意义(P <0. 05)。地塞米松组降低LTB4含量的效果优于哮喘宁颗粒组,差异有统计学意义(P <0. 05)。布地奈德组降低LTB4含量的效果优于哮喘宁颗粒组,差异有统计学意义(P <0. 05)。哮喘宁颗粒组、布地奈德组、地塞米松组大鼠血清CRP的表达降低程度相当,差异无统计学意义(P> 0. 05)。结论哮喘宁颗粒能下调哮喘大鼠LTB4、LTC4、CRP的表达,其治疗哮喘的机制可能是通过抑制气道相关炎症介质来实现的。
Objective To observe the regulative effect of xiaochuanning granules on the expressions of LTB4,LTC4 and CRP in the asthma model rats and explore the impacts of xiaochuanning granules on the airway related inflammatory mediators in asthma rats so as to provide the laboratory evidences in the treatment with xiaochuanning granules.Methods A total of male Sprague-Dawley rats were randomized into 5 groups,named a blank control group,a model group,a xiaochuanning granules group,a budesonide group and a dexametasone group,12 rats in each one.The sensitization with ovalbumin and intraperitoneal injection with aluminium hydroxide and ovalubumin nasal stimulation were adopted to prepare asthma models.The double sandwich ELISA was applied to determine the changes of the expressions of LTB4 and LTC4 in lung tissue homogenate of rats in each group.The speed scattering turbidimetric assay was used to determine the expressions of CRP.Results In the model group,the expressions of LTB4,LTC4 and CRP in lung tissue were increased,indicating the significant differences as compared with the blank control group(P<0.05).After treatment,the expressions of these indexes in the xiaochuanning granules group,the budesonide group and the dexametasone group were all reduced,indicating the significant differences as compared with the model group(P<0.05).The decrease of LTB4 in the dexametasone group was superior to the xiaochuanning granules group,indicating the significant difference(P<0.05)and the decrease in the budesonide group was better than the xiaochuanning granules group,indicating the significant difference(P<0.05).The decreases of CRP expression in the xiaochuanning granules group,the budesonide group and the dexametasone group were similar,without significant differences in comparison(P >0.05).Conclusion Xiaochuanning granules down-regulate the expressions of LTB4,LTC4 and CRP in asthma rats and the effect mechanism is probably through the suppression of airway related inflammatory mediators.
