两台荧光定量PCR仪检测HBV DNA结果的一致性分析

Analyse the consistency of HBV DNA results detected by two fluorescent quantitative PCR instruments

目的:评价ABI Vii A 7 Dx和ABI 7300实时荧光定量PCR仪检测HBV DNA结果的一致性并分析仪器间结果的偏倚。方法:按照美国临床实验室标准化委员会EP9-A2标准的要求,以通过卫生部室间质量评价的ABI 7300实时荧光定量PCR仪作为参比仪器,ABI Vii A 7 Dx实时荧光定量PCR仪作为实验仪器。分别在两台PCR仪上检测40份血浆标本的HBV DNA,对两台PCR仪的检测结果进行比对并计算判断偏倚。结果:ABI Vii A 7 Dx和ABI 7300实时荧光定量PCR仪检测HBV DNA结果间的相关性较好(r=0. 983 5),两台PCR仪检测结果的偏倚可以接受。结论:ABI Vii A7 Dx和ABI 7300实时荧光定量PCR仪检测HBV DNA结果一致性较好,可以同时用于临床HBV DNA的检测。

Objective:To evaluate the consistency of HBV DNA results detected by ABI ViiA 7 Dx and ABI 7300 fluorescent quantitative PCR instrument and analyse the suitability of the results between the instruments.Methods:According to NCCLS EP9-A2 standard,ABI 7300 fluorescent quantitative PCR instrument passed the inter-room quality evaluation of the Ministry of health was used as the reference instrument and ABI ViiA 7 Dx fluorescent quantitative PCR instrument was used as the test instrument.HBV DNA of 40 plasma samples were detected by the two PCR instruments,respectively.The results of the two instruments were compared and the bias was calculated.Results:There was a good correlation between the results of HBV DNA detected by the two PCR instruments,and the bias of the results was acceptable.Conclusion:The consistency of HBV DNA results detected by ABI ViiA 7 Dx and ABI 7300 fluorescent quantitative PCR instrument is good.The two instruments can be used in clinical at the same time.
