美洲大蠊提取物对肝硬化门静脉高压症大鼠VEGF的影响 被引量:1

摘要 目的观察美洲大蠊提取物(APA)对门静脉高压症大鼠血管内皮生长因子(VEGF)的影响。方法采用CCl4加乙醇制作大鼠门静脉高压症模型,造模及药物干预9周后用直接测压法测各组大鼠门静脉压力,同时用免疫组化法检测胃组织VEGF水平,并观察肝脏组织形态学改变。结果模型组门静脉压力较正常组明显升高(P<0.01);APA组及心得安组静脉压力较正常组也明显升高(P<0.01),但较模型组明显下降(P<0.01)。APA1、APA2组胃组织中VEGF的表达明显下降(P<0.01),且受损肝脏的病理形态明显改善。结论 APA不仅可以改善门静脉系统的高动力状态,其治疗效果优于心得安,而且还能降低胃组织中VEGF的表达和改善受损肝脏的病理形态,在肝硬化门静脉高压症治疗中的作用值得进一步研究。 Objective To investigate the effects of extract of Periplaneta americana(APA)on the vascular endothelial growth factor(VEGF)in rats with portal hypertension.Methods The rat portal hypertension models was prepared by adopting CCl4 and alcohol.After 9 weeks,and the portal vein pressure in each group was measured by using the direct manometric method.The expression level of VEGF in gastric tissue was detected by immunohistochemistry,and the liver tissue morphology changes were observed.Results The portal vein pressure of the model group was significantly increased compared with the normal group(P<0.01),the venous pressure in the APA group and propranolol group was significantly increased compared with the normal group(P<0.01),but was significantly decreased compared with the model group(P<0.01);the expression of gastic tissue VEGF in the APA1 and APA2 groups was decreased,moreover the pathological morphology of injured liver was significantly improved.Conclusion The APA not only can improve the hyperdynamic status of portal system,its treatment effect is superior to that of propranolol,moreover reduces the VEGF expression in gastric tissue and improves the pathomorphism of injured liver,its role in treating portal hypertension of liver cirrhosis deserves further study.
