啤酒活性干酵母凝血质提取研究

摘要 [目的]研究并优化啤酒活性干酵母凝血质的提取工艺。[方法]生长曲线法确定其培养时间,采用L_9(3~4)正交试验设计,应用低温和葡聚糖凝胶(SephadexG-75)对凝血质进行纯化。[结果]啤酒活性干酵母最佳培养时间为9 d,凝血质提取优化工艺参数为乙醇浓度85%、提取温度35℃、第1次丙酮体积(丙酮∶粗提液)为0.7、恒温磁力搅拌器转数为600 r/min,凝血质提取量可以达到1 000 mL活化液0.660 g,纯化率为13%。[结论]从啤酒活性干酵母可提取凝血质,方法简单易实现,降低凝血质动物来源的生成成本,具有极大的应用价值。 [Objective]The research aimed to study and optimize the extraction technology of clotting substance from active dry yeast of beer.[Method]The growth curve method was used to determine the culture time.The L 9(3^4)orthogonal test was used to purify the blood coagulation using a low temperature and Sephadex G-75.[Result]The optimal culture time of beer active dry yeast was 9 d,the optimum technological parameters for coagulation extraction were 85%of ethanol concentration,the extraction temperature was 35℃,the first acetone volume(acetone∶crude extract)was 0.7,and the constant temperature magnetic mixing device was 600 r/min.The amount of coagulant extraction could reach 0.660 g of 1 000 mL active liquid,purification rate was 13%.[Conclusion]Dried yeast from beer activity can be extracted from clotting matter,the method is simple and easy to realize,reducing the cost of clotting animal source,and has great application value.
