辽宁凤城市红松中松材线虫的分离与鉴定 被引量:22

摘要 近年来松材线虫Bursaphelenchus xylophilus在我国由南向北逐渐扩张并危害新的寄主。对辽宁省凤城市样地中红松Pinus koraiensis受害情况调查,利用显微镜形态观察和分子测序的方法对病原线虫进行综合鉴定。结果发现:凤城市样地中半枯和枯死红松体内均存在松材线虫,且87.7%以上为扩散型3龄幼虫,其分子鉴定结果与NCBI数据库中松材线虫的ITS和28S多个序列均完全相同,综合判断枯死红松中的病原线虫为松材线虫,由此确定松材线虫在我国自然条件下侵染红松。 In recent years,pine wood nematode has gradually expanded from south to north in China and harmed new hosts.The occurrence of pine wilt disease in the samples of Pinus koraiensis in Fengcheng,Liaoning Province was detected.Nematodes isolated from dead P.koraiensis were also identified by microscopic observation and molecular sequencing.The results showed that pine wood nematode was found in the P.koraiensis with yellowish needles,and more than 87.7%pine wood nematodes were third-stage dispersal juveniles.The result of molecular identification indicated that the ITS and 28S sequences of pine wood nematode identified were same as the sequences in the NCBI database.Through comprehensive judgment,the pathogenic nematode of dead P.koraiensis was pine wood nematode.The pine wood nematode began to invade P.koraiensis under natural conditions in China.
