摘要
目的探讨Kiss-1和AEG-1在胃癌组织中的表达及其相关性。方法采用RT-PCR和免疫组织化学法检测77例胃癌组织和20例正常胃组织中Kiss-1和AEG-1的表达,进一步用Western blot方法检测Kiss-1和AEG-1在胃癌组织和正常胃组织中表达。结果 RT-PCR结果发现胃癌组织中Kiss-1 mRNA表达水平显著低于正常胃组织(P<0.05),AEG-1mRNA表达水平显著高于正常胃组织(P <0. 05);免疫组化和Western blot发现胃癌组织中Kiss-1蛋白表达水平显著低于正常胃组织(P <0.05),AEG-1蛋白表达水平显著高于正常胃组织(P <0.05);Kiss-1 mRNA及蛋白阳性表达与胃癌组织的浸润深度、淋巴结转移有关(P<0.05);AEG-1 mRNA及蛋白阳性表达与胃癌组织的分化程度有关(P <0.05);胃癌组织中Kiss-1和AEG-1的表达呈负相关。结论 Kiss-1在胃癌组织中表达降低,AEG-1在胃癌组织中表达升高,Kiss-1、AEG-1可能参与胃癌的发生、发展以及浸润并有望成为诊断胃癌的生物学标记。
Objective To explore the expressions and clinical significances of Kiss-1 and AEG-1 in the tissues of gastric carcinoma. Methods The expressions of Kiss-1 and AEG-1 in 77 cases of gastric carcinoma and 20 cases of normal gastric were detected by RT-PCR,immunohistochemically staining method and western blot analysis. Results RT-PCR showed that the mRNA expression of Kiss-1 was significantly higher in the tissues of gastric carcinoma than normal gastric tissues,while AEG-1 showed the opposite result.Immunohistochemically staining method and western blot analysis revealed that Kiss-1 was decreased in the gastric carcinoma tissues compared with normal gastric tissues.However,the expression of AEG-1 protein was upregulated in gastric carcinoma tissues.The positive expression rate of Kiss-1 was closely correlated with invasion depth and lymph node metastasis and AEG-1 was closely correlated with differentiation degree.Spearman’s correlation coefficient showed the negative correlation of the content of Kiss-1 in gastric carcinoma tissues and AEG-1 in normal gastric tissues. Conclusion Kiss-1 in the gastric carcinoma tissues showed a low expression compared with normal gastric tissues,whereas a contrary trend was observed for AEG-1.Kiss-1 and AEG-1 might play important roles in the occurrence and development of gastric cancer and could be used as biological markers for the diagnosis of gastric carcinoma.
