酶法辅助提取甘蔗渣中水溶性多糖的工艺优化 被引量:11

摘要 通过实验筛选出果胶酶作为甘蔗渣中水溶性粗多糖提取率最高的一种酶,对酶解条件进行单因素试验和正交试验优化,得到酶法辅助提取甘蔗渣中水溶性多糖的最佳工艺条件:酶解温度50℃,酶解pH 5.0,酶用量2.0%,酶解时间50 min。在此提取条件下做验证试验,甘蔗渣粗多糖的提取率为1.09%±0.011%,比推荐的较佳酶解条件提高了10.4%,达到了预期的优化效果。通过对酶法辅助提取到的甘蔗渣粗多糖溶液进行紫外扫描,在260~280nm处均无明显吸收峰,进一步说明了该提取工艺条件的可靠性。 The best extraction rate of water-soluble crude polysaccharides from bagasse is screened by experiments.The optimum enzymatic hydrolysis conditions are optimized by single factor test and orthogonal test.The optimum conditions for the extraction of water-soluble polysaccharides from bagasse are as follows:the enzyme hydrolysis temperature is 50℃,the enzymatic pH is 5.0,the enzyme dosage is 2%and the enzymatic time is 50 min.The extraction rate of crude polysaccharides from bagasse is 1.09%±0.011%,which increases by 10.4%compared with the recommended enzymatic hydrolysis conditions,and achieves the desired optimization effect.There is no obvious absorption peak at 260~280 nm by UV scanning of the crude polysaccharides solution extracted from the bagasse,and the reliability of the extraction conditions is further explained.
