摘要
分别在291K,306K和323K温度下利用荧光光谱法对柠檬黄和溶菌酶的相互作用进行了探讨。结果显示,柠檬黄能使溶菌酶发生荧光猝灭,此过程为静态猝灭。测定了柠檬黄与溶菌酶反应体系的结合常数和结合位点数。利用该体系反应过程的热力学参数,确定了柠檬黄与溶菌酶体系的作用力类型。由非辐射能量转移理论,计算得到体系的结合距离。并采用同步荧光法和三维荧光法考察了柠檬黄对溶菌酶构象的影响。
The interaction between lemon yellow and lysozyme was studied by fluorescence spectrometry at 291KL 306K and 323K,respectively.The results showed that the fluorescence quenching of lysozyme could be realized by the process of static quenching.The binding constants and binding sites of the reaction system were determined.Based on the thermodynamic parameters of the reaction process,the interaction type of the system was determined.Based on the theory of non radiative energy transfer,the binding distance of the system is calculated.The effect of citric acid on lysozyme conformation was investigated by synchronous fluorescence and three-dimensional fluorescence.
