一株蛴螬绿僵菌的分子鉴定

Molecular Identification of MetarhiziumIsolated from Cadaver of White Grub

以形态特征为主要依据的传统分类鉴定方法,难以区分绿僵菌属内隐秘种,而DNA分子鉴定能更加准确的对绿僵菌属隐秘种的分类地位进行鉴别。为明确一株分离自蛴螬的绿僵菌Ma1518菌株的具体分类归属,我们通过对该菌株的rDNA-ITS序列、β-tubulin序列和RPB2序列进行PCR扩增、测序及系统进化树构建,明确其为平沙绿僵菌Metarhizium pinghaense。

In order to confirm the taxonomic status of a Metarhizium that isolated from the cadaver of white grub.Amplification,sequencing,and system evolution tree construction of thenuclear ribosomal internal transcribed spacer(rDNA-ITS),β-tubulin and RNA polymerase II second largest subunit(RPB2)geneof the fungusMa1518were carried out.The result showed that the entomopathogenic fungus was identified as Metarhizium pinghaense.
