利用TCGA数据库分析GRB2在肝细胞肝癌中的表达及临床意义

Analysis of GRB2 expression in hepatocellular carcinoma and its clinical significance using TCGA database

目的分析GRB2在肝细胞肝癌(HCC)中的表达与HCC患者临床特征及预后的相关性,进而预测GRB2与HCC发生发展的关系。方法从癌症基因组图谱(TCGA)中选取HCC中GRB2的基因表达谱及相关临床和病理资料,分析GRB2在HCC癌组织和癌旁组织中的表达情况以及HCC癌组织中GRB2表达量与HCC患者临床特征及预后的关系。结果 GRB2在HCC癌组织中的表达高于癌旁组织(P<0.05)。GRB2表达水平与HCC临床病理分级、TNM分期、血清甲胎蛋白(AFP)水平相关,HCC临床病理分级、TNM分期、血清AFP水平越高,GRB2表达量越高(P<0.05)。COX多因素分析显示GRB2高表达和TNM分期是影响HCC患者预后的独立危险因素。结论 GRB2可作为促癌基因在HCC患者的肿瘤组织中呈高表达,提示HCC患者预后不良,有望成为早期诊断HCC的分子标志物和判断预后的指标。

Objective To analyze the correlation between the expression of GRB2 in hepatocellular carcinoma(HCC)and clinical features and prognosis of patients with HCC,further to predict the relationship between GRB2 and development of HCC.Methods The gene expression profile of GRB2 in HCC and related clinical and pathological data were selected from the cancer genome atlas(TCGA).The GRB2 expression in HCC cancer tissues and adjacent tissues was analyzed,and the relationship between GRB2 expression and clinical characteristics and prognosis of patients with HCC was analyzed.Results The expression of GRB2 in HCC cancer tissues was higher than that in adjacent tissues(P<0.05).The expression level of GRB2 was correlated with clinical pathological grade of HCC,TNM stage,and serum alpha-fetoprotein(AFP)levels.The higher the clinical pathological grade,TNM stage and serum AFP level of HCC were,the higher the expression of GRB2 was(P<0.05).COX multivariate analysis showed that the TNM stage and over expression of GRB2 were the independent risk factors affecting prognosis of patients with HCC.Conclusion GRB2 can be used as a oncogene in patients with HCC with high expression in cancer tissue.It suggests that the prognosis of patients with HCC is poor.GRB2 is expected to be a molecular marker for early diagnosis and a prognostic indicator of HCC.
