油茶枯黄酮类化学成分及其体外抗炎活性 被引量:15

Chemical constituents of the flavonoids from Camellia oleifera and their antiinflammatory activities in vitro

摘要 目的研究油茶枯Camellia oleifera Abel Cake中黄酮类化学成分及其体外抗炎活性。方法油茶枯80%乙醇提取物采用硅胶、HPLC、中低压制备色谱进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用脂多糖(LPS)诱导小鼠巨噬细胞RAW264. 7建立体外炎症筛选模型评价其抗炎活性。结果从中分离得到7个化合物,分别鉴定为山柰酚(1)、槲皮素(2)、芦丁(3)、山柰酚-3-O-β-D-吡喃葡萄糖(1→2)-α-L-吡喃阿拉伯糖苷(4)、山柰酚-3-O-α-L-鼠李糖(1→6)-β-D-吡喃葡萄糖苷(5)、山柰酚-3-O-[2-O-β-D-木糖-6-O-α-L-鼠李糖]-β-D-葡萄糖苷(6)、山柰酚-3-O-[2-O-β-D-半乳糖-6-O-α-L-鼠李糖]-β-D-葡萄糖苷(7)。化合物1,4~7对LPS诱导的RAW264. 7细胞产生的炎症因子NO、PGE2均具有良好的抑制作用,且均能抑制RAW 264. 7细胞中NF-κB的活性、下调诱导型一氧化氮合酶(i NOS)和环氧合酶-2 (COX-2)的表达。结论化合物4为首次从该植物中分离得到;化合物1,4~7具有较强的体外抗炎活性。 AIM To study the chemical constituents of the flavonoids from Camellia oleifera Abel Cake and their anti-inflammatory activities in vitro.METHODS The 80%ethanol extract from C.oleifera was isolated and purified by silica column,HPLC and low and medium pressure chromatography,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their anti-inflammatory activities in vitro were estimated by LPS-induced inflammatory model in RAW264.7 macrophages.RESULTS Seven compounds were isolated and identified as kaempferol(1),quercetin(2),rutoside(3),kaempferol-3-O-β-D-glucopyranosyl(1→2)α-L-arabinopyranoside(4),kaempferol-3-O-α-L-rhamnopyranosyl(1→6)-β-D-glucopyranoside(5),kaempferol-3-O-[2-O-β-D-xylopyranosyl-6-O-α-L-rhamnopyranosyl]-β-D-glucopyranoside(6),kaempferol-3-O-[2-O-β-D-galactopyranosyl-6-O-α-L-rhamnopyranosyl]-β-D-glucopyranoside(7).Compounds1,4-7 had very significant inhibitory effects on inflammatory mediators such as NO and PGE2,and could inhibit the activity of NF-κB in RAW 264.7 cells and reduced the expression of inducible nitric oxide synthase(i NOS)and cyclooxygenase-2(COX-2).CONCLUSION Compound 4 is isolated from this plant for the first time,and compounds 1,4-7 have strong anti-inflammatory activities in vitro.
