利拉鲁肽对动脉粥样硬化过程中糖尿病大鼠血管平滑肌KCa3.1表达的影响 被引量:1

摘要 目的探讨利拉鲁肽对动脉粥样硬化过程中糖尿病大鼠血管平滑肌KCa3. 1表达的影响。方法 SD大鼠24只,分为对照组6只,模型组9只,治疗组9只。模型组与治疗组大鼠给予高脂饲料喂养,空白对照组大鼠给予普通饲料,共4周;模型组与治疗组腹腔注射STZ 30 mg/kg,空白对照组注射等体积柠檬酸钠缓冲液;造模成功后治疗组使用利拉鲁肽0. 3 mg/kg 1次/d皮下注射,对照组、模型组大鼠皮下注射等容积生理盐水,共11周。处死大鼠后对大鼠主动脉血管进行HE染色、Massion三色染色,Real-time RT-PCR进行血管平滑肌KCa3. 1mRNA定量测定,Westernblotting测定血管平滑肌蛋白表达。结果模型组和治疗组均造糖尿病模型成功。HE染色提示模型组及治疗组有早期的动脉粥样硬化,HE可见治疗组及对照组血管平滑肌增殖及紊乱,Massion三色提示模型组及治疗组有胶原纤维增生。Realtime-PCR测定表明,模型组主动脉平滑肌细胞KCa3. 1mRNA表达比治疗组及对照组高(P <0. 05),治疗组及对照组无显著统计学差异(P> 0. 05)。Western-blotting测定表明,模型组主动脉平滑肌细胞KCa3. 1蛋白表达比治疗组及对照组高(P <0. 05),治疗组及对照组无显著统计学差异(P> 0. 05)。结论利拉鲁肽能够对抗早期动脉粥样硬化,减低糖尿病大鼠血管平滑肌KCa3. 1表达。 Objective To explore effect of liraglutide on the expression of KCa3.1 in vascular smooth muscle of diabetic rats during atherosclerosis.Methods 24 SD rats were divided into control group,model group,and treatment group.The model group and the treatment group were fed with high fat diet,and in the blank control group were given normal feed for 4 weeks.The model group and the treatment group were intraperitoneally injected with STZ 30 mg/kg,and the blank control group was injected with an equal volume of sodium citrate buffer.After successful modeling,the treatment group was treated with liraglutide 0.3 mg/kg once daily.Rats in the control group and the model group were injected subcutaneously with an equal volume of normal saline for 11 weeks.After the rats of three groups were put to death,HE staining,Massion staining,Real-time PCR and Western-blotting were used to determine the expression of KCa3.1 mRNA and protein in vascular smooth muscle.Results Diabetic rats of the model group and the treatment group were established successfully.HE staining indicated that the model group and the treatment group had early atherosclerosis.HE showed the proliferation and disorder of vascular smooth muscle in the treatment group and the model group.Massion trichrome staining suggested that the model group and the treatment group had collagen fiber hyperplasia.Realtime-PCR assay showed that KCa3.1 mRNA in aortic smooth muscle cells in model group was higher than that in treatment group and control group(P﹤0.05).KCa3.1 mRNA had no significant statistic difference between treatment group and control group(P﹥0.05).Western-blotting assay showed that KCa3.1 protein in aortic smooth muscle cells in model group was higher than those in treatment group and control group(P﹤0.05).KCa3.1 protein had no significant statistic difference between treatment group and control group(P﹥0.05).Conclusion Liraglutide can resist early atherosclerosis and decrease the expression of KCa3.1 in vascular smooth muscle of diabetic rats.
